Pengaruh Kombinasi Ekstrak Pegagan (Centella asiatica L.) dan Jahe (Zingiber officinale) dalam Induksi Lipolisis Jaringan Adiposa pada Tikus Wistar Betina berdasar Parameter Histologi

Kulit merupakan bagian terluar manusia yang berpotensi mengalami beberapa masalah yang menyebabkan penampilan menjadi kurang menarik. Salah satu masalah estetika berkaitan dengan kulit yang umumnya dialami para wanita adalah selulit. Selulit merupakan perubahan penampakan pada kulit yang meyerupai kulit jeruk (orange-peel). Ekstrak pegagan dan jahe secara terpisah dilaporkan mampu memperbaiki kondisi kulit akibat selulit. Namun, efek campuran kedua ekstrak tersebut belum diketahui. Penelitian ini bertujuan untuk mengetahui kemampuan sediaan uji campuran ekstrak pegagan dan jahe dalam induksi lipolisis yang ditunjukkan dengan kemampuan penurunan ukuran diameter sel lemak pada tikus Wistar betina yang dipapar diet lemak tinggi. Digunakan tikus betina galur Wistar berjumlah 25 ekor tikus yang dibagi menjadi 5 kelompok tikus, yaitu: kelompok kontrol normal (Na-CMC 0,5%), kontrol negatif (lemak babi), kelompok dosis 1, 2, dan 3 masing-masing 50, 100, dan 200 mg/kg BB. Semua kelompok hewan uji diberi lemak babi kecuali kontrol normal mulai hari ke-1 hingga hari ke-30. Sediaan uji yang diberikan dalam bentuk nanopartikel yang sebelumnya dilarutkan dalam Na-CMC dan pemejanan dilakukan secara peroral. Pada hari ke-30, hewan uji dikorbankan dan diambil kulitnya untuk dianalisis secara histologi pada jaringan adiposa. Seluruh data dianalisis dengan One Way ANOVA dengan taraf kepercayaan 95%. Data yang diperoleh menunjukkan bahwa kombinasi ekstrak pegagan dan jahe dapat memperkecil diamater sel lemak secara signifikan seiring bertambahnya dosis (dose dependent), sehingga dapat ditarik kesimpulan jika kombinasi ekstrak tersebut dapat menginduksi lipolisis pada jaringan adiposa dengan memperkecil diameter sel lemak.

Skin is the outermost part of human who might experience some problems that cause less attractive appearance. One of the aesthetic problems associated with the skin that are generally experienced by women is cellulite. Cellulite is a change in the appearance of the skin that resemble an orange peel. Centella asiatica extract and ginger separately reported to improve the condition of the skin due to cellulite. However, the effects of the extract mixture is unknown. This study aims to determine the ability of the test preparation gotu kola extract and ginger mixture in the induction of lipolysis which is indicated by the ability of the fat cells decrease in size of the diameter of the female Wistar rats were exposed to a high fat diet. Used female Wistar rats amount to 25 rats were divided into 5 groups namely: normal control group (Na-CMC 0,5%), negative control (lard), dose group 1, 2, and 3 respectively 50, 100 and 200 mg/kg body weight. All groups of test animals were given lard except the normal control from day 1 to day 30. The test preparation is given in the form of nanoparticles which previously dissolved in Na-CMC by oral administration. On the 30th day, the test animals were sacrificed and their skins were analyzed histologically for adipose tissue. All data were analyzed by One Way ANOVA with 95% confidence level. Data obtained showed that the combination of gotu kola extract and ginger can reduce fat cell diameter significantly with increasing dose (dose-dependent), so it can be concluded if the combination of the extract can induce lipolysis in adipose tissue by reducing the diameter of the fat cells.
