Hyperlipidemia increase free radicals production and causing oxidative stress. This oxidative stress can affect lipid profiles such as cholesterol and triglycerides, as well as the levels of antioxidant enzymes such as SOD, CAT, and POD. Oxidative stress can also affects the expression of the Heme Oxygenase gene. Neoagarooligosaccharide (NAOS) is an agarose hydrolyzed from red algae and has antihyperlipidemic and antioxidant properties. Therefore, this study was conducted to test the antihyperlipidemic and antioxidant potentials of NAOS enzymatically hydrolyzed from red algae in hyperlipidemic mice (Mus musculus Linnaeus, 1758). In this study, mice undergo HFD and STZ injection to induces hyperlipidemia. The treatments carried out in this study were metformin and NAOS doses of 100, 200, and 400 mg/kgbw. Blood samples were then taken for lipid profiles (cholesterol and triglycerides) and antioxidant enzymes (SOD, CAT, and POD) analysis. While the liver was used to measure hmox1 gene expression. The results were then analyzed using the GraphPad application. NAOS administration can lower cholesterol and triglyceride levels, as well as improve the activity of antioxidant enzymes, especially CAT level, which is significantly increased by NAOS. Furthermore, NAOS administration also induces hmox1 gene expression, thus enable hepatocytes to fight free radicals and reduce oxidative stress due to HFD. The results obtained from this study are expected to increase the insight of the public and researchers regarding the use and benefits of the antioxidant properties of NAOS in combating oxidative stress and treating hyperlipidemia.
