EFEK WAKTU KONTAK CU-ZEOLIT ALAM KONSENTRASI 0,8 MOLAR DALAM KEMASAN PAPER WRAPPED SEBAGAI BAHAN DESINFEKTAN TERHADAP PERTUMBUHAN BAKTERI STREPTOCOCCUS PYOGENES ATCC 19165 \ DAN PSEUDOMONAS AERUGINOSA ATCC 27853

Cu-zeolit merupakan material yang mampu menekan pertumbuhan bakteri dengan karakteristik cation reversible, molecular sieve dan adsorpsi dimana kondisi tersebut dapat dikembangkan menjadi desinfektan non toksik terhadap manusia tanpa mengurangi inaktivasi mikroorganisme. Tujuan: Mengetahui efek waktu kontak Cu-zeolit alam 0,8 M kemasan paper wrapped pada rendaman aquades terhadap pertumbuhan bakteri S.pyogenes dan P.aeruginosa . Metode penelitian : penelitian eksperimental dengan simple randomized design. Sebanyak 10 gram Cu-zeolit dikemas dalam kantong kertas Whatmann no 42 berukuran 5x5 cm, dikontakkan selama 15,30 dan 45 menit dalam 99,5 ml akuades yang terpapar 0,5x108 CFU/ml S.pyogenes dan 0,5x108 CFU/ml P.aeruginosa. Selanjutnya dihitung pertumbuhan bakteri menggunakan metode Total Plate Count. Hasil rerata pertumbuhan bakteri S.pyogenes paparan menit ke 15, 30 dan 45 (1840 �± 571,236 CFU ; 29 �± 16,33 CFU dan 0 CFU ) sedangkan P.aeruginosa berturut-turut (2776 �± 725,277 CFU; 55 �± 23,214 CFU dan 0 CFU). Uji t-test Cu-zeolit terhadap pertumbuhan bakteri lama kontak menit ke 15 dan 30 antara S.pyogenes terhadap P.aeruginosa berbeda bermakna(p=0,005) sedangkan Uji Mann-Whitney kontak menit ke 45 tidak bermakna (p=1). Hasil uji Mann-Whitney Cu-zeolit terhadap kontrol positif pada menit ke 15 dan 30 berbeda bermakna (p= 0,001), sedangkan hasil uji kontak menit 45 menunjukkan perbedaan tidak bermakna (p=1,00).Kesimpulan: semakin lama waktu kontak Cu-zeolit alam konsentrasi 0,8 M kemasan paper wrapped maka semakin besar penurunan pertumbuhan bakteri S. pyogenes dan P.aeruginosa, dimana penurunan pertumbuhan bakteri S.pyogenes lebih besar dibanding P.aeruginosa kontak menit ke 15 dan 30 serta mampu membunuh total bakteri S.pyogenes dan P.aeruginosa kontak menit ke 45.
Infection of a surgical wound due to bacteria is a major problem for surgical patients. Cu-zeolite is a material that can suppress bacterial growth with cation reversible characteristics, molecular sieve and adsorption to be developed into non-toxic disinfectants to humans without reducing the inactivation of microorganisms. Objective: to observe the effects of 0.8 M contact time of natural Cu-zeolite on paper wrapped packaging in aquadest immersion on the growth of S.pyogenes and P.aeruginosa bacteria. Methods: experimental research with simple randomized design. 10 grams of Cu-zeolite were packaged in Whatman no 42 paper bags measuring 5 x 5 cm, contacted for 15, 30 and 45 minutes in 99.5 ml of distilled water exposed to 0.5 x 108 CFU / ml of S.pyogenes and 0.5x108 CFU / ml P .aeruginosa. Subsequently, bacterial growth was calculated with the Total Plate Count method. The average growth of S.pyogenes bacteria was exposed to 15, 30 and 45 minutes (1840 �± 571,236 CFU; 29 �± 16.33 CFU and 0 CFU) while P.aeruginosa was (2776 �± 725,277 CFU; 55 �± 23,214 CFU and 0 CFU) respectively. Based on the t-test on Cu-zeolite, the bacterial growth in the 15th and 30th minute contact between S.pyogenes to P.aeruginosa was significantly different (p = 0.005) while on the basis of Mann-Whitney test, in the 45th minute contact was not significant (p = 1). Based on Mann-Whitney test to Cu-zeolite on positive controls in the 15th and 30th minute contacts were significantly different (p = 0.001), while the results of the 45th minute contact test showed no significant differences (p = 1.00).Conclusion: the longer the contact time of natural Cu-zeolite concentration of 0.8 M paper wrapped packaging, the greater the decrease in growth of S. pyogenes and P.aeruginosa bacteria, where the decrease in S.pyogenes bacterial growth was greater than that of P.aeruginosa contact in the 15th and 30th minute and was able to kill the total S.pyogenes bacteria and P.aeruginosa in the 45th minute contact time.
