PENGARUH KONSENTRASI AIR MEDIUM PERTUMBUHAN TERHADAP PRODUKSI LIPID KAPANG OLEAGINOUS ISOLAT BR 2.2

Kapang oleaginous merupakan salah satu mikroorganisme yang mampu mengakumulasikan banyak biomassa dengan cepat (dalam 96-130 jam) dan kerap digunakan dalam produksi lipid. Pertumbuhan kapang sangat bergantung pada komposisi kimia yang ada pada lingkungan pertumbuhannya. Medium pertumbuhan yang digunakan pada kapang harus mengandung karbohidrat tinggi sebagai sumber nutrient dan kandungan nitrogen yang tinggi pula. Salah satu sumber karbon yang dapat digunakan kapang dalam proses pertumbuhan adalah glukosa. Isolat BR 2.2 merupakan kapang oleaginous yang mampu mengakumulasi 28.44% lipid dari total biomassa keringnya dengan glukosa sebagai sumber karbon dan dalam volume medium 50 mL. Oleh karena itu, penelitian ini dilakukan untuk mengetahui pengaruh variasi volume air medum dan waktu inkubasi terhadap produksi biomassa serta lipid isolate BR 2.2. Produksi biomassa dan lipid dianalisis pada volume 10, 20, 30, 40, dan 50 mL dengan waktu inkubasi 48, 96, dan 144 jam. Hasil penelitian menunjukkan bahwa akumulasi lipid dan produksi biomassa meningkat seiring dengan pengurangan kadar air pada medium pertumbuhan dan mencapai angka tertinggi pada volume medium 20 mL dengan waktu inkubasi 144 jam, yakni 0,87±0,04 g/L dan 12,53±0,29 g/L. Sedangkan penyerapan glukosa oleh kapang meningkat seiring pengurangan kadar air pada medium sementara penyerapan nitrogen menurun seiring pengurangan air pada medium. Oleaginous fungi is one of the microorganisms that can accumulate a lot of biomass quickly (within 96-130 hours) and is often used in the production of lipids. The growth of fungi is very dependent on the chemical composition of the environment in which it grows. The growth medium of fungi must contain high carbohydrates as a source of nutrients and high nitrogen content as well. One of the carbon sources that can be used by fungi in the growth process is glucose. BR 2.2 isolate is an oleaginous fungi that capable of accumulating 28.44% lipids from the total dry biomass with glucose as carbon source in 50 mL of growth medium. Therefore, this study was conducted to determine the effect of variations in the volume of medium and incubation time on the production of biomass and lipid isolate BR 2.2. Biomass and lipid production were analyzed at volumes of 10, 20, 30, 40, and 50 mL with incubation times of 48, 96, and 144 hours. The results showed that lipid accumulation and biomass production increased with the reduction of water content in the growth medium and reached the highest number in the medium volume of 20 mL with an incubation time of 144 hours, i.e. 0.87±0.04 g/L and 12.53±0.29 g/L. Meanwhile, the absorption of glucose by the fungi increased with the reduction of water content in the medium, while the absorption of nitrogen decreased with the reduction of water in the medium.
