EFEKTIVITAS PENGGUNAAN MINERALIZED DAN DEMINERALIZED GOAT CORTICAL BONE XENOGRAFT PADA FRAKTUR FEMUR KELINCI

Efektivitas Penggunaan Mineralized dan Demineralized Goat Cortical Bone Xenograft Pada Fraktur Femur Kelinci Sherli Melani Ludji 19/448631/PKH/00716 INTISARI Fraktur merupakan kondisi terputus atau rusaknya kontinuitas jaringan tulang, baik karena trauma secara langsung maupun tidak langsung. Tingkat kejadian fraktur dapat dibantu dengan pemberian bahan cangkok untuk mengganti fragmen patahan tulang, sehingga dapat merangsang proses kesembuhan tulang. Penelitian ini bertujuan untuk mempelajari efektifitas penggunaan mineralized dan demineralized goat cortical bone xenograft pada fraktur femur kelinci. Pembuatan bahan cangkok dengan menggunakan tulang diafisis kambing. Proses pembuatan bahan cangkok meliputi bahan cangkok mineralisasi dan demineralisasi. 24 ekor kelinci jantan umur 3-4 bulan, berat badan 3-5 kg yang digunakan dalam penelitian ini dibagi menjadi 3 kelompok dengan masing-masing kelompok berjumlah 8 ekor kelinci. Kelompok I adalah kelinci yang dipergunakan sebagai kontrol, pada diafisis tulang femur kiri dibor dengan diameter 0,5 cm, tanpa pemberian bahan cangkok, kelompok II adalah kelinci yang pada diafisis tulang femur kiri dibor dengan diameter 0,5 cm dan diberi bahan cangkok MGCBX, kelompok III adalah kelinci yang pada diafisis tulang femur kiri dibor dengan diameter 0,5 cm dan diberi bahan cangkok DGCBX. Monitoring perkembangan kesembuhan dilakukan pada minggu ke-2 dan minggu ke-6 pasca operasi dengan pemeriksaan jumlah total leukosit, pemeriksaan foto rontgent, dan biopsi tulang femur. Data perhitungan total leukosit dianalisis dengan Univariate Analysis of Variance pola faktorial menunjukkan tidak ada perbedaan jumlah total leukosit diantara ketiga kelompok (P>0,05), namun waktu pengambilan sampel berpengaruh terhadap jumlah total leukosit (P<0,05). Analisis radiologis menunjukkan pada minggu ke-6 pasca operasi kelompok kontrol masih tampak kalus dan belum terjadi mineralisasi, kelompok MGCBX dan DGCBX memperlihatkan penyatuan fragmen tulang kembali ke bentuk normal, masa kalus yang terbentuk sudah mengalami mineralisasi dan bahan cangkok sudah terdegradasi secara sempurna. Hasil analisis histopatologi menunjukkan pada minggu ke-4 kelompok kontrol terjadi reaksi inflamasi dan pada minggu ke-6 pasca operasi masih terdapat gap fraktur. Pada semua kelompok terjadi proses kesembuhan tulang ditandai dengan terbentuknya tulang trabekula, proliferasi jaringan ikat, sel osteogenik dan vaskularisasi baru. Berdasarkan hasil penelitian, maka dapat diambil kesimpulan bahwa MGCBX dan DGCBX dapat dipergunakan sebagai bahan cangkok alternatif untuk penanganan fraktur pada kelinci.

The Efficacy of Mineralized Goat Cortical Bone Xenograft and Demineralized Goat Cortical Bone Xenograft in Femur Fracture in Rabbit Sherli Melani Ludji 19/448631/PKH/00716 ABSTRACT Fracture is a condition of disconnection or damage to the continuity of bone tissue, either due to direct or indirect trauma. The rate of fracture incidence can be assisted by using the provision of graft materials to replace bone fracture, which stimulates the bone-healing process. This study aims to learn the effectiveness of the mineralized and demineralized goat cortical bone xenograft fracture femur utilization in the rabbit. The construction of graft material uses the goat diaphysis bone. The process of making graft materials includes mineralized and demineralized graft materials. The 24 male rabbits aged 3-4 months, the bodyweight 3-5 kg used in this study are divided into 3 groups with 8 rabbits each. Group I contains the rabbits that are used as the control, which is on the left femur diaphysis is drilled with a diameter of 0.5 cm, without grafting, group II contains the rabbits, which are drilled on the left femur diaphysis with a diameter of 0.5 cm and provided with MGCBX graft material, Group III contains the rabbits, which on the left femur diaphysis is drilled with a diameter of 0.5 cm and provided with DGCBX graft material. The recovery progress monitoring is held on the second week and sixth week postoperatively by examining the total number of leukocytes, examining X-rays, and biopsy of the femur. The data for calculating the total leukocytes, which are analyzed by two-way ANOVA shows that there was no difference in the total number of leukocytes among these three groups (P> 0.05), but the time of sampling affects the total number of leukocytes (P <0.05). Radiological analysis shows that at the sixth week after surgery, the control group still shows callus and there is no mineralization, the MGCBX and DGCBX groups show that the bone fragments are reunited to their normal form, the reformed callus mass has undergone mineralization, and the graft material has completely degraded. The results of the histopathological analysis show that in the fourth week the control group had an inflammatory reaction and in the sixth week postoperatively, there is still a fracture gap. In all groups, there is a bone healing process characterized by the formation of trabecular bone, proliferation of connective tissue, osteogenic cells, and new vascularity. Based on the research results, it could be concluded that MGCBX and DGCBX can be used as alternative graft materials for fracture management in rabbits.
