Pengaruh Suplementasi Kalsitriol Pada Tikus Wistar Ovariektomi Terhadap Gambaran Histopatologis Uterus

2016 | Skripsi | S1 KEDOKTERAN HEWANPenelitian ini dilakukan untuk mengetahui pengaruh suplementasi kalsitriol terhadap gambaran histopatologis uterus tikus wistar ovariektomi. Lima belas ekor tikus Wistar betina umur 8 minggu dibagi menjadi tiga kelompok, yaitu kelompok kontrol non-ovariektomi (NK), kelompok kontrol ovariektomi (OVK) dan kelompok ovariektmi + kalsitriol (OVD) masing-masing kelompok terdiri dari 5 ekor. Tikus kelompok NK dan OVK diberi pakan standar, sedangkan kelompok OVD diberi pakan standar dan suplemen kalsitriol dosis 100ng/hari/tikus selama dua bulan. Pada akhir perlakuan tikus dieutanasi dan uterus tikus diambil dan difiksasi dalam formalin 10% untuk pemeriksaan histopatologis dengan pewarnaan Hematoxylin-Eosin. Hasil pemeriksaan histopatologis uterus tikus NK tidak terdapat perubahan pada lapisan epitelium endometrium maupun stroma endometrium. Uterus tikus OVK mengalami pemendekan epitelium menjadi kuboid simpleks hingga skuamus simpleks dan penurunan jumlah glandula uterina pada stroma endometrium. Epitelium endometrium uterus tikus OVD berbentuk kuboid simpleks hingga kolumner simpleks, menyerupai bentuk normal epitelium. Jumlah glandula uterina bertambah dan stroma endometrium rapat. Dari hasil penelitian dapat disimpulkan bahwa pemberian suplemen kalsitriol dosis 100ng/hari/tikus dapat meningkatkan jumlah glandula uteria dan menyebabkan bentuk epitelium endometrium mendekati normal.
This research was aimed to investigate the effect of calcitriol supplementation on histopathological feature of uterine in ovariectomized Wistar rat. Fifteen female Wistar rat at 8 weeks of age were divided into three groups, that was non-ovariectomized control group (NK), ovariectomized group (OVK) and ovariectomized+calcitriol group (OVD), each group contain 5 rats. Rats of NK and OVK group were fed with standard diet, while OVD group rats were fed with standard diet and 100ng/day/rat calcitriol supplement for two months. At the end of treatment, rats were eutanized, the uterine was removed and fixated in formalin 10% for histopathological examination with Hematoxylin-Eosin staining. As the result of histopathological examination, there was no endometrial epithelium and endometrial stroma change of NK rat’s uterine. Endometrial epithelium of OVK rat’s uterine was shorthened into single layer of cuboidal to squamous and decrease in number of uterine gland in endometrial stroma. Endometrial epithelium of OVD rat’s uterine was single layer of cuboidal to columner, like its normal shape. The amount of uterine gland was increase and endometrial stroma was dense. It can be conclude that calcitriol supplementation 100ng/day/rat can increase in number of uterine glands and change the shape of endometrial epithelium like normal.
