TOKSISITAS SUBKRONIS PRODUK GD PADA TIKUS JANTAN WISTAR DENGAN PARAMETER HEMATOLOGI

Produk GD merupakan produk herbal yang diproduksi oleh produsen yang bekerjasama dengan Fakultas Farmasi UGM. Produk GD terdiri dari 50 mg ekstrak herba sambiloto dan 100 mg ekstrak daun mimba yang memiliki efek dalam membantu menurunkan kadar gula darah. Tujuan penelitian ini adalah untuk mengetahui gejala klinis dan efek toksik yang ditimbulkan akibat pemejanan bahan uji secara berulang selama 90 hari serta untuk mengetahui pengaruh pemejanan bahan uji secara berulang terhadap parameter hematologi pada hewan uji selama masa perlakuan (hari ke-90) dan masa reversibilitas (hari ke-118). Metode penelitian ini mengacu pada metode OECD 408 tentang uji toksisitas subkronis. Hewan uji yang digunakan dalam penelitian ini adalah tikus putih galur Wistar sebanyak 50 ekor tikus jantan, dikelompokkan menjadi 4 kelompok yaitu kelompok kontrol, Dosis I (40,5 mg/kgBB), Dosis II (202,5 mg/kgBB) dan Dosis III (1012,5 mg/kgBB). Data-data yang diperoleh seperti purata kenaikan berat badan per hari (PKBP) dan hasil pemeriksaan hematolgi selanjutnya dianalisis statistik menggunakan metode One Way ANOVA dan paired sample t-test dengan program SPSS 19. Hasil penelitian menunjukkan bahwa pemberian sediaan uji secara berulang selama 90 hari perlakuan terhadap tikus jantan galur Wistar dan 28 hari masa reversibilitas secara umum tidak menimbulkan gejala toksik, tidak mempengaruhi asupan makanan dan minuman, serta parameter hematologi.

GD product is herbal product manufactured by producers who cooperated with the Faculty of Pharmacy UGM. GD product composed of 50 mg extract of sambiloto herbs and 100 mg extract of neem leaves that has an effect in helping to lower blood sugar levels. The purpose of this studi was to determine the clinical symptoms and toxic effect caused by treatment oral given of GD product repeatedly during 90 days and to determine the affect of GD product to hematological parameters during 90 treatment days and after reversibility ( day 118). This research method refers to method OECD 408 about toxicity subchronic. Experimental rats are used in this research the male rat Wistar strain as much as 50 male rat. Experimental rats devide into 4 groups, control groups, Dose I (40,5 mg/kgBB), Dose II (202,5 mg/kgBB), and Dose III (1012,5 mg/kgBB). The data are obtained as the mean weight gain per day and the result of examination hematology are analyzed statistic by method one way ANOVA and paired sample t-test at program of SPSS 19. The result showed that treatment GD product repeatedly during 90 days to male rats Wistar strain and 28 days reversibility generally does not cause toxic symptoms, does not affect the intake of food and drinks, and hematological parameters.
