TOKSISITAS SUBKRONIS PRODUK SKM TERHADAP TIKUS WISTAR JANTAN DENGAN PARAMETER HEMATOLOGI

Sediaan uji produk SKM berisi campuran 75 mg ekstrak herba seledri, 75 mg ekstrak daun kumis kucing dan 75 mg ekstrak buah mengkudu merupakan produk ekstrak herbal milik PT. Swayasa Prakarsa yang berkhasiat membantu menurunkan tekanan darah. Akan tetapi belum ada pengujian tentang keamanan penggunaan kombinasi ekstrak tersebut sehingga penelitian ini bertujuan untuk mengevaluasi toksisitas subkronis sediaan uji produk SKM pada tikus jantan galur Wistar terhadap parameter hematologi. Penelitian ini mengarah pada guideline OECD 408. Hewan uji yang digunakan sebanyak 50 ekor, dikelompokkan menjadi 3 kelompok peringkat dosis dan 1 kelompok kontrol. Pengelompokan terdiri atas kelompok kontrol (Na CMC 0,5%) sebanyak 15 ekor, dosis 1 (dosis 121,5 mg/kg BB) 10 ekor, dosis 2 (dosis 364,5 mg/kg BB) 10 ekor dan dosis 3 (dosis 1093,5 mg/kg BB) sebanyak 15 ekor. Peringkat dosis diperoleh berdasarkan penggunaan SKM pada manusia (bobot 70 kg) yaitu 3 kali sehari 1 kapsul atau sebesar 1350 mg perhari dikalikan faktor konversi dari manusia 70 kg terhadap tikus dengan bobot 200 g yaitu 0,018. Pengamatan yang dilakukan meliputi gejala klinis, perkembangan berat badan, asupan makanan dan minuman, serta analisis hematologi. Hasil penelitian menunjukkan bahwa pemberian sediaan uji produk SKM peroral selama 90 hari pada tikus jantan galur Wistar terbukti tidak menimbulkan gejala-gejala klinis dan kematian, tidak mempengaruhi asupan makanan dan minuman, namun pemberian sediaan uji SKM selama 90 hari mempengaruhi PKBP dari hewan uji pada dosis 364,5 mg/kgBB, mempengaruhi parameter hematologi pada nilai MCV yaitu dosis 121,5 mg/kgBB, dosis 364,5 mg/kgBB dan dosis 1093,5 mg/kgBB serta efek bersifat terbalikkan. Test preparation of SKM product containing a mixture of 75 mg celery extracts, 75 mg of kumis kucing leaf extract and 75 mg of noni fruit extract is a herbal extract product produced by PT. Swayasa Prakarsa, it is efficacious to help lower blood pressure. However, there has been no testing on safety of the combined use of the aforementioned extracts. Hence, this study aims to evaluate the sub-chronic toxicity test preparation of SKM products on Wistar male trhough hematological parameters. This research based on the OECD 408 guideline. Animal tests used were 50 animals, divided into three groups based on the dose ranked and 1 control group. Grouping consisted of the control group (Na CMC 0,5%) as 15 animals, dose 1 (dose of 121.5 mg / kg body weight) 10 animals, dose 2 (dose of 364.5 mg / kg body weight) 10 animals and dose 3 (dose of 1093.5 mg / kg body weight) 15 animals. Ranked dose was obtained by the use of SKM on humans (weight of 70 kg), 1 capsule 3 times daily or 1350 mg per day multiplied by the conversion factor of human, 70 kg, towards mice with a weight of 200 g, which was 0,018. Observations were conducted on the clinical symptoms, weight development, food and beverage intake, as well as hematological analysis. The results showed that the giving of the test preparation of SKM products orally for 90 days to Wistar male rats proven to not cause clinical symptoms and death, it did not affect the intake of food and beverages. However, the giving of the test preparation of SKM product for 90 days affected: PKBP of the test animals at doses of 364.5 mg / kg body weight; hematology parameters at the MCV value from a dose of 121.5 mg / kg body weight, a dose of 364.5 mg / kg body weight and a dose of 1093.5 mg / kg body weight and the effects were irreversible.
