OPTIMASI PROPILEN GLIKOL DAN AVICEL PH 200 PADA FORMULA TABLET LIKUISOLID GLIBENKLAMID

Kecepatan disolusi glibenklamid menjadi laju penentu absorbsi. Metode untuk meningkatkan kecepatan disolusi obat dapat dilakukan dengan pembuatan tablet likuisolid. Penelitian ini bertujuan untuk mengetahui formula optimum sediaan tablet likuisolid glibenklamid menggunakan propilen glikol dan Avicel PH 200 serta mengetahui pengaruh propilen glikol dan Avicel PH 200 terhadap sifat fisik tablet. Sebanyak delapan run dengan variasi kadar propilen glikol dan Avicel PH 200 dibuat menggunakan Design Expert 8.0.7.1 trial. Evaluasi yang dilakukan antara lain uji sudut diam, uji keseragaman bobot, uji kekerasan, uji kerapuhan, uji waktu hancur, dan uji disolusi. Penetapan formula optimum tablet likuisolid glibenklamid dilakukan menggunakan metode simplex lattice design. Hasil percobaan menunjukkan formula optimum diperoleh pada proporsi propilen glikol sebanyak 30 mg dan Avicel PH 200 sebanyak 240 mg. Hasil uji sudut diam, kekerasan, kerapuhan, dan waktu hancur dari formula optimum tidak berbeda bermakna dengan prediksi.
Glibenclamide is type two diabetes drug which has poor solubility and it is classified in Biopharmaceitical Classification System II. Dissolution becomes rate limiting step of glibenclamide absorption. Enhancing dissolution of drugs can be done by making liquisolid tablet. The aim of this study is to know optimum formula of glibenclamide liquisolid tablet using propylene glycol and Avicel PH 200 and to know the effect of propylene glycol and Avicel PH 200 towards physical properties of glibenclamid liquisolid tablet. As many as eight runs with variation amount of propylene glycol and Avicel PH 200 were made by Design Expert 8.0.7.1 trial. Evaluations that were conducted include angle of repose, weight uniformity, tablets hardness, friability, tablet disintegration, and dissolution test. Determination of optimum formula was conducted using simplex lattice design method. The results of this study showed that optimum formula was obtained at proportion of propylene glycol as many as 30 mg and Avicel PH 200 as many as 240 mg. The result of angle of repose test, hardness test, friability test, and disintegration test were not significantlly different with the predictions.
