EFEK INDUKSI HIPOTIROID DENGAN PROPILTHYOURACIL (PTU) TERHADAP KADAR HORMON TRIIODOTIRONIN (T3) PADA TIKUS PUTIH (Rattus norvegicus)

Hipotiroid merupakan kondisi terganggungnya fungsi kelenjar tiroid sehingga kadar hormon tiroid dalam darah baik dalam bentuk Triiodothyronin (T3) maupun Thyroxin (T4) terlalu rendah. Propylthiouracil (PTU) merupakan obat yang sering digunakan untuk induksi hipotiroid dengan indikator naiknya kadar Thyroid Stimulating Hormone (TSH). Penelitian ini bertujuan mengetahui pengaruh pemberian Propylthiouracil terhadap kadar triiodotironin tikus wistar. Penelitian ini menggunakan 42 ekor tikus jantan berumur 3-4 bulan. Tikus diadaptasikan selama satu minggu sebelum diberi perlakuan. Pemberian Propylthiouracil (PTU) dilakukan secara peroral selama dua minggu dengan dosis 54 mg/kg. Darah tikus diambil sebelum perlakuan dan dua minggu setelah perlakuan. Hasil penelitian menunjukkan bahwa tikus yang diinduksi menggunakan Propylthiouracil (PTU) selama dua minggu mengalami kenaikan kadar Triiodothyronin (T3) yang signifikan (P<0,05). Kesimpulan yang dapat diambil, bahwa pemberian Propylthiouracil (PTU) untuk induksi hipotiroid dengan dosis 54 mg/kg selama dua minggu untuk induksi hipotiroid belum mampu menurunkan kadar Triiodothyronin (T3) pada tikus.

Hypothyroidism is a disfunction of the thyroid glandthat caused decreasing of the thyroid hormone both Triiodothyronin (T3) and Thyroxin (T4) in the blood. Propylthiouracil(PTU) is an antithyroid drug, used widely to increase the Thyroid Stimulating Hormone (TSH) level as a hypothyroidism indicator. The aim of this study is to determine the level of Triiodothyronin in Wistar rat induced by Propylthiouracil. This study used 42 male ratsaged 3-4 months, divided into two groups. After adapted for a week, Propylthiouracil (PTU) given at a dose 54 mg/kg body weight, administrated orally for two weeks. The rat's blood was taken before treatment and two weeks after treatment. The results showed significant increased level of Triiodothyronin (T3) (P<0,05). The conclusion is the administration of Propylthiouracil (PTU) for the hypothyroidism induction at a dose of 54 mg / kg for two weeks have not been able to decrease Triiodothyronin (T3) levels of rats.
