PROFIL HEMATOLOGIS TIKUS PUTIH (Rattus norvegicus Berkenhout, 1769) WISTAR DISLIPIDEMIA DENGAN PEMBERIAN AIR PERASAN BUAH LUWINGAN (Ficus hispida L.f.) PER ORAL

Konsumsi makanan tinggi lemak dan rendah serat, menyebabkan abnormalitas pada kadar lipid dalam darah dan mampu memicu timbulnya dislipidemia. Dislipidemia ditandai dengan kenaikan kadar kolesterol total, LDL, trigliserida serta penurunan kadar HDL dalam darah. Dislipidemia dapat merusak membran sel pada sel darah yang akan mengganggu keseimbangan kerja komponen selular darah. Buah Ficus hispida L.f (luwingan) mengandung senyawa flavonoid dan saponin yang diduga mampu menurunkan kadar lipid dalam darah. Darah merupakan komponen penting dalam tubuh dan pemeriksaan darah rutin dapat digunakan untuk menunjang diagnosa suatu penyakit dan melihat respon fisiologis tubuh terhadap suatu penyakit. Penelitian ini bertujuan untuk mempelajari profil hematologis tikus (Rattus norvegicus Berkenhout, 1769) betina galur Wistar dislipidemia dengan pemberian air perasan buah luwingan (Ficus hispida L.f.) per oral. Pada penelitian ini digunakan 15 ekor tikus yang dibagi menjadi 5 kelompok yaitu kelompok Kontrol (K), tikus dislipidemia + air perasan buah luwingan mentah (DFMen), tikus + air perasan buah luwingan matang (DFMat), tikus dislipidemia + obat simvastatin (DObat), tikus dislipidemia + pergantian diet pakan basal (DPB). Penelitian dilakukan selama 56 hari dengan pemberian asupan tinggi lipid pada hari ke-0 hingga ke-56 serta pemberian air perasan buah luwingan mentah, matang dan obat simvastatin pada hari ke-28 hingga ke-56. Pemeriksaan profil hematologis (jumlah total eritrosit, kadar hemoglobin, hematokrit, jumlah total leukosit, jumlah total limfosit, jumlah total neutrofil, rasio N/L dan jumlah total trombosit) dilakukan menggunakan Hematology Analyzer Sysmex pada hari ke 0; 28; 35; 42; 49; 56. Hasil penelitian di analisis dengan uji one way ANOVA kemudian dilanjutkan dengan Uji Duncan Multiple Range Test (DMRT) menggunakan SPSS v 16.0. Hasil penelitian menunjukkan bahwa pemberian air perasaan buah luwingan matang pada tikus dislipidemia secara nyata mampu menjaga kualitas profil eritrosit, leukosit dan trombosit tetap pada kisaran normal. Namun air perasan buah luwingan mentah dapat menurunkan profil eritrosit dan trombosit dibawah kisaran normal

Consumption of high fat and low fiber foods, causing abnormalities in blood lipid levels and can trigger of dyslipidemia. Dyslipidemia characterized by increased levels of total cholesterol, LDL, triglycerides and decreased levels of HDL. Dyslipidemia can damage cell membrane of blood cells and disrupt their work. Ficus hispida L.f (luwingan) fruits contains flavonoids and saponins, that were suspected able to lowering blood lipid levels. Blood is an important component in body. Routine blood tests can be used to diagnose of disease and observe the physiological response of body. The aim of this research is to study the hematological profile of (Rattus norvegicus Berkenhout, 1769) dyslipidemic female Wistar rats after oral administration of luwingan (Ficus hispida L.f.) fruits filtrate per oral. This study used 15 rats were divided into 5 groups: control (K), dyslipidemic rats+unripe luwingan fruits filtrate (DFme), dyslipidemic rats+ripe luwingan fruits filtarte (DFma), dyslipidemic rats+simvastatin (DObat), dyslipidemic rats+basal diet (DPB). The study was conducted for 56 days by giving a high lipids on day 0 until 56 th. On the 28th day until the 56th was giving unripe and ripe luwingan fruits filtrate and drug simvastatin. Examination of hematological profile (total number of erythrocytes, hemoglobin concentration, hematocrit, total number of leukocytes, total lymphocyte count, the total number of neutrophils, the ratio of N/L and the total number of platelets) by using Hematology Analyzer Sysmex on days 0; 28; 35; 42; 49; 56 th. The results analyzed by one way ANOVA test followed by Duncan's Multiple Range Test Test (DMRT) using SPSS v 16.0. The results showed that giving ripe luwingan fruits filtrate in dyslipidemic rats significantly able to maintain the quality profile of erythrocytes, leukocytes and thrombocytes in the baseline. But unripe luwingan fruits filtrate was lowering profile of erythrocytes and leukocytes exceeds the baseline.
