TOXICITY AND ANTIBACTERIAL ACTIVITY SCREENINGS OF EXTRACT OF MARINE SPONGE Theonella sp. COLLECTED FROM BARRANG LOMPO ISLAND INDONESIA

Penapisan toksisitas dan aktivitas antibaktteri ekstrak spons Theonella sp. dari Pulau Barrang Lompo Indonesia telah dilakukan. Ekstraksi sampel spons dilakukan dengan diklorometana/metanol (2:1). Ekstrak diklorometana yang telah dipekatkan dipartisi dalam etil asetat/air (1:1) kemudian ekstrak etil asetat yang diperoleh dipisahkan dengan kromatografi kolom. Ekstrak etil asetat dan fraksi-fraksi hasil pemisahan kolom kromatografi diuji toksisitas dan aktivitas antibakteri dengan metode Brine Shrimp Lethality Test (BSLT) dan Metode difusi agar. Hasil uji BSLT pada ekstrak etil asetat dan fraksi 7 menunjukkan toksisitas tinggi dengan harga LC50 masing-masing 0,160 dan 2.463 micro gram/mL. Fraksi- fraksi menunjukkan aktivitas antibakteri lemah dengan zone diameter penghambatan maksimum 12 mm. Analisis dengan LC-ESI/MS mengindikasikan fraksi 7 mengandung empat senyawa yaitu: rifamycin S, reticulatine B,theonellapeptolide Ib dan swinholide J. Kata kunci: Theonella, toksisitas, antibakteri

Toxicity and antibacterial activity screenings of the extract of marine sponge Theonella sp. collected from Barrang Lompo Island Indonesia have been conducted. The sponge sample was extracted using dichloromethane/methanol (2:1). The dichloromethane layer was evaporated and partitioned by ethyl acetate/water (1:1) and was subjected to column chromatography. Brine Shrimp Lethality Test (BSLT) toxicity and antibacterial agar diffusion assays were the biological screening methods of the ethyl acetate crude extract and column chromatography fractions. The BSLT showed high toxicities against Artemia salina of that the crude ethyl acetate extract and fraction 7 with LC50 values of 0.160 and 2.463 micro gram/ml, respectively. The column chromatography fractions of the ethyl acetate extract showed weak antibacterial activities with maximum of zone of inhibition diameter of 12 mm. The LC-ESI/MS analysis of fraction 7 indicated that there are four known compounds: rifamycin S, reticulatine B, theonellapeptolide Ib and swinholide J.
