KEMAMPUAN REGENERASI DAN STRUKTUR HASIL REGENERASI SIRIP KAUDAL IKAN WADER PARI (Rasbora lateristriata Bleeker, 1854) NORMAL DAN CACAT SIRIP

Ikan wader sangat ekonomis sebagai kuliner dengan cita rasa lezat. Permintaan ikan tersebut meningkat drastis, namun ketersediaan di alam menurun. Budidaya wader bergantung pada kualitas lingkungan yang sangat penting untuk pertumbuhan dan perkembangan ikan. Sirip kaudal adalah bagian tubuh ikan yang rentan rusak oleh faktor eksternal padahal fungsinya sangat penting untuk kehidupan ikan. Penelitian aspek biologis wader sangat penting untuk mendukung budidaya ikan tersebut. Tujuan penelitian ini untuk mempelajari kemampuan regenerasi sirip kaudal ikan wader pari (Rasbora lateristriata Bleeker, 1854). Ikan diperoleh dari Laboratorium Struktur dan Perkembangan Hewan Fakultas Biologi, UGM. Jumlah total ikan 36 ekor berumur 4 bulan, dengan panjang 4-5 cm, ikan dikelompokkan dalam ikan normal dan ikan cacat sirip kaudal. Bagian pangkal sirip kaudal ikan dipotong, lalu dipelihara selama 4 minggu. Setelah 4 minggu, ikan dikorbankan, kemudian diambil kaudanya lalu dibuat preparat histologis dengan pewarna Hematoxylin Eosin dan Mallory asam Fuchsin serta preparat anatomis tulang kauda dengan pewarna Alizarin's Red Alcian Blue, dilanjutkan pengamatan menggunakan mikroskop cahaya. Data kuantitatif diperoleh dengan pengukuran panjang sirip kaudal hasil regenerasi setiap 2 hari dan data kualitatif diperoleh dari pengamatan histologis dan anatomis kauda ikan. Hasil menunjukkan bahwa ikan normal dan ikan cacat sebagian bisa meregenerasi sirip kaudal, namun ikan dengan cacat seluruh kaudanya tidak dapat beregenerasi. Pengamatan histologis menunjukkan bahwa ada perbedaan struktur jaringan ikat pada ikan normal dan ikan cacat. Pengamatan anatomi tulang kauda menunjukkan perbedaan kekakuan sirip hasil regenerasi dengan sirip normal tanpa pemotongan. Kesimpulan dari studi ini terdapat perbedaan kemampuan regenerasi sirip kaudal ikan normal dan ikan cacat, serta terdapat perbedaan struktur histologis dan anatomis hasil regenerasi dari sirip normal dan sirip tanpa pemotongan.

Waders has high economical value as a culinary fish with delicious taste. The demand for fish availibility and supply are increasing drastically, nowdays. The cultivation of waders compare to the quality of environment are very important for the growth and development of fish. Caudal fin are the body parts of fish that susceptible of the damage caused by external factors whereas its functions are very important in the survival of the fish. The study on biological aspects of the wader is very important to support the cultivation.This research aim to study the caudal fins regeneration ability of yellow rasbora (Rasbora lateristriata Bleeker, 1854). Fish were obtained from Laboratory of Animal Structure and Development Faculty of Biology, UGM. Total number of 36 four months old fishes, with length of 4-5 cm, were grouped into the normal fish and defected fish, for fishes each. The proximal caudal fins of fish are cut and then kept for 4 weeks. After 4 weeks, the fish were sacrificed, and then the caudal were processed for histological preparation and stained with Hematoxylin-Eosin and Mallory Acid Fuchsin and for the anatomical sceleton preparations of cauda with Alizarin's Red-Alcian Blue staining, the next process is to observe it using the light microscope. The quantitative data is obtained by the measurement of the caudal fins length which are the results of the regeneration once of 2 days and the qualitative data is obtained by the histological observation and anatomical caudal fin. The results showed that the normal and the partial defect fish could regenerate the caudal fin, however the fish with no caudal fin couldnot regenerate. Histological observations indicate that there were differences of the structure of the connective tissue between normal and defect fish. Anatomical observation of the sceleton caudal showed a difference of the stiffnes with the normal fins without ampution. The conclusion of this study were caudal fin regeneration ability rate and histological structure on normal fish and defect fish, and then there is a difference in the histological and anatomical structure of the regeneration caudal fin of normal fins and without amputation.
