STRUKTUR HISTOLOGIS REN, DAN KADAR KREATININ, UREUM, ASAM URAT MENCIT (Mus musculus Linnaeus, 1758) JANTAN PADA TOKSISITAS AKUT ETIL ASETAT (PER ORAL)

Etil asetat adalah senyawa organik berwujud cair, tidak berwarna, memiliki aroma khas dan diproduksi dalam sklala besar sebagai pelarut plastik, pembersih cat kuku, dan perisa makanan. Etil asetat diketahui dapat menyebabkan sakit kepala, kulit kering, kekurangan darah, kerusakan organ, peradangan pada mata, dan saluran pernapasan pada dosis tertentu. Penelitian ini bertujuan untuk mengetahui pengaruh etil asetat terhadap struktur ren, kadar kreatinin, ureum, dan asam urat mencit (Mus musculus Linnaeus, 1758) jantan. Penelitian ini dilakukan pada 45 ekor mencit (Mus musculus Linnaeus, 1758) jantan galur swiss dengan bobot antara 22-26 g usia 6-7 minggu. Mencit dibagi menjadi 5 kelompok yaitu kontrol (tanpa cekok aquades), kontrol (dengan cekok aquades), dosis 0,2622 g/kg BB; 0,7868 g/kg BB; dan 1,049 g/kg BB. Dosis diberikan secara oral pada hari ke-0 dan dilakukan pengamatan struktur histologis, pengukuran kadar kreatinin, ureum, dan asam urat pada hari ke 5, 10, dan 15. Pembuatan preparat histologis dilakukan menggunakan metode parafin dengan pewarnaan Haematoxylin-Eosin (HE) dan pengujian kadar kreatinin, ureum dan asam urat dengan metode spektrofotometrik enzimatik. Hasil penelitian pada semua dosis hari ke-5 sampai hari ke-15 merubah struktur histologis ren dan tidak mempengaruhi kadar pada kreatinin, ureum, dan asam urat.

Ethyl acetate is liquid organic compound, colorless, has distinctive smell and are produced in large scale as plastic solvents, nail polish remover, and fruit flavor. Ethyl acetate is known to cause headaches, dry skin, lack of blood, organ damage, inflammation of the eyes, and respiratory tract at certain doses. Experiment to observe the effects of ethyl acetate on renal structure and measurement of creatinin, ureum, and uric acid of the male mice (Mus musculus Linnaeus, 1758) have been conducted. Fourty five Swiss�¢ï¿½ï¿½ male mice (Mus musculus Linnaeus, 1758) by weight of 22-26 g and of ranged age of 6 -7 weeks have been selected. These male mices are divided into five groups, namely group with aquades, group without aquades, group of given dose of 0,2622 g/kg Wt, group of given dose of 0,7868 g/kg Wt, and group of given dose of 1,049 g/kg Wt. The doses were given on the zeroth day. Observation on histological structure and measurement of creatinin, ureum and uric acid were conducted on the days of 5th, 10th and 15th, respectively. Histological preparation has been prepared using parafin method with Haematoxylin-Eosin (HE) stain. Measurement of creatinin, ureum, and uric acid were performed using spectrofotometric-enzymatic method. Resuls of observation on the days of 5th until the day of 15th show that there are not changed on renal histological structure and not affect on of creatinin, ureum, and uric acid levels.
