THE EFFECT OF BOVINE VITREOUS GEL IN STIMULATING PROLIFERATION OF HUMAN DERMAL SENESCENT FIBROBLASTS

Fibroblast tua, yang tidak responsif terhadap rangsangan TGF- Beta 1, gagal untuk berkembang biak. Meningkatkan kekuatan mikro-mekanis dengan asam hialuronat dapat memperbaiki jalur sinyal TGF- Beta 1 fibroblast penuaan manusia. Sebagai sampah dari rumah pemotongan hewan, sapi-vitreous dianggap sebagai sumber kadar asam hyaluronic tinggi. Tujuan penelitian: Untuk mengevaluasi efek dari sapivitreous gel pada artifisial berusia fibroblast dermal manusia dalam: tarif proliferasi in vitro Bahan dan metode: fibroblas dermal Manusia dari enam manusia yang berbeda akan dirangsang untuk menjadi fibroblas tua karena kelaparan serum. Setelah itu, mereka akan diperlakukan dengan berbagai konsentrasi sapi yang vitreous diencerkan dalam DMEM + 1% janin bovine serum selama 72 jam. Proliferasi seluler akan dibandingkan dan dianalisis. Data akan disajikan dalam mean plus minus standar error dan dianalisis dengan ANOVA atau Friedman tergantung pada normalitas distribusi data. Hasil: Biakan sel fibroblas tua dalam vitreous gel mata sapi 50% dan 25% menunjukkan peningkatan indeks proliferasi secara bermakna (p<0.05). Indeks proliferasi fibroblast tua dalam vitreous gel mata sapi 50% dan 25% adalah: 5.09 dan 5.08. Backgrounds: The most responsible factor in poor healing of chronic ulcers is wound bed fibroblasts aging. The aged fibroblasts, which are unresponsiveness against TGF- Beta 1 stimulation, fail to proliferate. Increasing microenvironment-mechanical forces with hyaluronic acid may ameliorate TGF- Beta 1 signaling pathway of human aging fibroblasts. As trash of slaughterhouses, bovine-vitreous is considered as a source of high hyaluronic acid levels. Aim of study: To evaluate effects of bovine-vitreous gel on artificially aged human dermal fibroblast in: proliferation rates in vitro Materials and methods: Human dermal fibroblasts from six different human foreskins will be stimulated to become senescent fibroblasts by serum starvation. After that, they will be treated with various concentrations of bovine's vitreous diluted in a DMEM + 1% fetal bovine serum for 72 hours. The cellular proliferations will be compared and analyzed. Data will be presented in mean plus minus standard error and analyzed with ANOVA or Friedman depends on normality of data distributions. Result: Cultivation of senescent fibroblast in 50% and 25% bovine vitreous gel showed significant (p<0.05)elevation in proliferation index. Proliferation index of senescent fibroblast in 50% and 25% bovine vitreous gel were 5.09 and 5.08 respectively. Conclusion : Bovine vitreous gel could increase the proliferation of senescent human dermal fibroblast.
