Potensi Antiviral Protein Tinta 70 kDa dari Kelinci Laut (Aplysia dactylomela, Rang 1828) terhadap Virus Avian Influenza Subtipe H5N1 Kiki Sakina Fakultas Biologi Universitas Gadjah Mada, kikisakina92@gmail.com INTISARI Virus Avian Influenza adalah virus penyebab penyakit avian influenza (AI) atau di Indonesia dikenal dengan istilah flu burung. Virus AI, terutama Highly Pathogenic Avian Influenza (HPAI) subtipe H5N1 dikategorikan sebagai penyakit zoonosis yang berbahaya. Kelinci laut merupakan organisme laut yang tergolong dalam moluska opisthobranchia. Kelinci laut dilaporkan mampu mensekresikan tinta yang mengandung protein antiviral. Tujuan dari penelitian ini adalah untuk mengetahui potensi antiviral protein tinta 70 kDa dari kelinci laut (Aplysia dactylomela, Rang 1828) terhadap virus AI dengan uji hemaglutinasi (HA) dan Real Time Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR). Skrining potensi antiviral protein tinta 70 kDa dari kelinci laut (Aplysia dactylomela, Rang 1828) terhadap virus AI dilaksanakan di Laboratorium Virologi Balai Besar Veteriner (BBVet) Wates, Yogyakarta menggunakan uji in ovo. Virus AI yang digunakan adalah koleksi Balai Besar Veteriner (BBVet) Wates Yogyakarta dengan nama A/duck/Kulon Progo/04141074-06/2014 clade 2.3.2. Telur ayam bertunas SAN (specific antibody negative) umur 9 hari dibagi menjadi 5 kelompok perlakuan (n = 3). Kelompok 1 sampai 3 diinokulasi dengan virus AI yang diberi perlakuan protein tinta 70 kDa dengan konsentrasi 10, 20 dan 40 mikrogram/mL, secara berturut-turut. Kelompok yang tidak diberi perlakuan digunakan sebagai kontrol negatif dan kelompok yang diinokulasi virus AI dengan pelarut digunakan sebagai kontrol positif. Uji hemaglutinasi digunakan untuk menghitung unit virus AI. Hasil uji real time RT-PCR menunjukkan bahwa nilai Cycle Threshold (Ct) protein 70 kDa konsentrasi 40 mikrogram/mL sama dengan nilai Ct kontrol. Hasil penelitian menunjukkan bahwa perlakuan protein tinta 70 kDa dari kelinci laut (Aplysia dactylomela, Rang 1828) tidak berpotensi sebagai antiviral terhadap virus AI subtipe H5N1 hingga konsentrasi 40 mikrogram/mL.

Antiviral Potency of 70 kDa Ink Protein from Sea Hare (Aplysia dactylomela, Rang 1828) Against H5N1 Subtype Avian Influenza Virus Kiki Sakina Faculty of Biology Gadjah Mada University, kikisakina92@gmail.com ABSTRACT Avian influenza virus is a virus caused a highly contagious disease of avian influenza or in Indonesia is known as flu burung. Avian influenza virus especially Highly Pathogenic Avian Influenza (HPAI) is classified as a dangerous zoonotic disease. Sea Hare is a marine organism belonging molusca ophistobranchia. Sea Hare enable to secret an ink containing the antiviral protein. The aim of this study was to determine the antiviral potency of 70 kDa ink protein from Sea Hare (Aplysia Dactylomela, Rang 1828) against AI virus using haemaglutination (HA) test and real time Reverse Transcriptase-Polymerase Chain Reaction (PCR) test. Screening the antiviral potency of 70 kDa ink protein from Sea Hare (Aplysia Dactylomela, Rang 1828) against AI virus was done using an in ovo assays. A/duck/Kulon Progo/04/141074-06/2014 clade 2.3.2. was AI virus strain collection from Balai Besar Veteriner (BBVet) Wates Yogyakarta that used in this experiment. Nine days old specific antibody negative (SAN) embryonated chicken eggs were devided into 5 treatment groups (n = 3). Group 1 to 3 were inoculated with AI virus H5N1 subtype treated with 70 kDa Sea Hare ink protein at concentration of 10, 20 dan 40 mikrogram/mL, respectively. Un-inoculated group saved as negative control and group inoculated with virus and diluents save as positive control. Haemagglutination test was used to quantify the amount of AI virus units. The result of real time RT-PCR test showed that the Cycle Threshold (Ct) value of 70 kDa Sea Hare ink protein at concentration of 40 mikrogram/mL was same with the Ct value of control positive. This study showed that treatment 70 kDa ink protein from Sea Hare (Aplysia Dactylomela, Rang 1828) was not potentially as antiviral against AI virus H5N1 subtype until concentration 40 mikrogram/mL.
