PENGARUH AKTIVASI PLATELETE RICH PLASMA DENGAN KOLAGEN TERHADAP MIGRASI SEL FIBROBLAS LIGAMEN PERIODONTAL

Terapi bedah periodontal merupakan prosedur regeneratif yang bertujuan memperbaiki struktur dan fungsi jaringan periodonsium sehingga cukup kuat untuk mendukung gigi. Growth factor yang mutlak dibutuhkan dalam proses regenerasi jaringan dapat di hasilkan dari PRP yang di aktivasi. Dalam penelitian ini di gunakan kolagen sebagai activator PRP. Tujuanya adalah untuk melihat pengaruh aktivasi PRP dengan kolagen terhadap migrasi sel fibroblas ligamen periodontal. Enam kelompok perlakuan terdiri dari kontrol, PRP, PRP diaktivasi kolagen selama 1,2 3 dan 7 hari diaplikasikan pada kelompok sel yang di kultur pada microplate 24 sumuran, masing masing kelompok 10 sumuran yang dilukai dengan pipet tip steril kemudian diinkubasi pada suhu 370 C dan di amati pada 12 jam , 24 jam, 48 jam dan 72 jam. Pada setiap sumuran diambil gambar menggunakan inverted microscope dengan perbesaran 4 kali lensa objective dan program optilab viewer yang dipasang pada lensa okuler. Persentase migrasi sel dihitung dengan program adobe photoshop. Data dianalisis dengan uji ANOVA dua jalur yang dilanjutkan dengan uji LSD dengan derajat kepercayaan 95%. Hasil menunjukkan adanya perbedaan yang bermakna antar waktu dan kelompok perlakuan. Terdapat perbedaan bermakna antara kelompok kontrol, PRP dan PRP yang di aktivasi kolagen. Kesimpulan penelitian ini adalah aktivasi PRP dengan kolagen dapat meningkatkan migrasi sel fibroblas pada ligament periodontal.

Treatment of periodontal surgery is regenerative procedures that aims to improve the structure and function of the periodontium tissue that is strong enough to support the teeth. Growth factor is absolutely necessary in the process of tissue regeneration can be generated from the PRP activation. In this study collagen was used as activator PRP. The aim of the study was to determine the effect of the to the migration of fibroblasts in the periodontal ligament. Six treatment groups consisted of control, PRP, collagen - activated PRP during 1,2 3 and 7 days applied to a group of cell cultures in 24 well microplate, each group composed 10 wells are wounded by a sterile pipette tip. Then it incubated at 370 C and observed at 12, 24, 48 and 72 hours. All wells were captured by using an inverted microscope with the objective lens magnification of 4 times and program optilab viewer program installed on the ocular lens. The percentage of cell migration was calculated by the program adobe photoshop. Data are analyzed by two way ANOVA followed by LSD test with confidence level of 95%. The result showed significant difference between time and treatment groups. There were significant difference between the control group, PRP and collagen - activated PRP. The conclusion of this study was the activation of collagen - activated PRP can improve the migration of fibroblasts in the periodontal ligament.
