FORMULASI SNEDDS (Self-Nano Emulsifying Drug Delivery System) EKSTRAK AKAR PURWOCENG GUNUNG DENGAN ASAM OLEAT

Purwoceng gunung adalah tumbuhan obat yang tumbuh di Indonesia dan diduga berkhasiat afrodisiak. Stigmasterol yang terdapat pada akar purwoceng gunung memiliki kelarutan yang rendah di dalam air, yakni sebesar 1, 12 x 10-5 mg/L. Penelitian ini bertujuan untuk memformulasikan ekstrak akar purwoceng gunung dalam bentuk SNEDDS (Self-Nano Emulsifying Drug Delivery System) sehingga kelarutannya dapat ditingkatkan. Akar purwoceng gunung diekstraksi dengan metode maserasi menggunakan etanol 70 %. Karakterisasi ekstrak akar purwoceng gunung meliputi kadar air, kadar abu, kadar stigmasterol, daya lekat, viskositas, dan rendemen. Formula SNEDDS ekstrak akar purwoceng gunung dibuat pada perbandingan 1:6:1 dengan komposisi asam oleat sebagai fase minyak, tween 20 sebagai surfaktan, dan propilen glikol sebagai kosurfaktan. Penentuan desain formula percobaan menggunakan metode simplex lattice design dengan bantuan program Design Expert versi 7.1.5. Respon kejernihan dan emulsification time dalam AGF digunakan untuk mengevaluasi formula SNEDDS. Karakterisasi formula optimum SNEDDS meliputi pengujian ukuran partikel dan distribusi ukuran partikel dengan PSA dan extract loading. Formula optimum SNEDDS ekstrak akar purwoceng gunung memiliki komposisi 90,614 % v/v tween 20 dan 9,386 % v/v propilen glikol. Nanoemulsi yang dihasilkan jernih dan homogen dengan nilai transmittan sebesar 94,9 % T± 0,73 , emulsification time dalam AGF sebesar 35,69 ± 4,34 detik, dan dapat menampung ekstrak sebesar 50 mg/mL.

Purwoceng gunung is a medicinal plant that grows in Indonesia and allegedly efficacious as aphrodisiac. Stigmasterol that contain in root of the purwoceng gunung have low solubility in water, which is equal to 1, 12 x 10-5 mg/L. This study aims to formulate purwoceng gunung root extract in the form of SNEDDS (Self-Nano Emulsifying Drug Delivery System) so that the solubility can be improved. Purwoceng gunung roots were extracted by maceration method using ethanol 70%. Characterization of the purwoceng gunung root extract include water content, ash content, content of stigmasterol, adhesion, viscosity, and yield. SNEDDS formula of purwoceng gunung root extract made at a ratio of 1: 6: 1 with the composition of oleic acid as an oil phase, tween 20 as surfactant, and propylene glycol as cosurfactant. Determination experimental design of SNEDDS formula using simplex lattice design method with the help of Design Expert version 7.1.5 program. Response of clarity and emulsification time in AGF (artificial gastric fluid) used to evaluate SNEDDS formula. Characterization of the optimum SNEDDS formula includes testing the particle size and distribution of particle size with PSA and extract loading. Optimum SNEDDS formula of the purwoceng gunung root extract has a composition of 90.614% v / v tween 20 and 9.386% v / v propylene glycol. The resulting nanoemulsion are clear and homogeneous with transmittan value amounted to 94.9% T ± 0.73, emulsification time in AGF amounted to 35.69 ± 4.34 seconds, and can accommodate extract at 50 mg / mL.
