2015 | Skripsi | S1 FARMASIEkstrak daun sirih (Piper betle L.) memiliki kandungan utama berupa eugenol yang memiliki aktivitas antibakteri yang bersifat iritan berat. Ekstrak diformulasikan dalam bentuk patch bukal mukoadhesif untuk memudahkan penggunaan dan kenyamanan pasien. Formula yang kontak lama dengan mukosa harus dievaluasi untuk memprediksi munculnya sensasi Stinging, Itching, Burning (SIB). Penelitian ini bertujuan untuk membuktikan munculnya sensasi SIB patch bukal mukoadhesif ekstrak daun sirih. Ekstrak daun sirih dibuat secara infundasi selanjutnya dilakukan proses freeze drying kemudian diuji spesifikasinya meliputi uji organoleptis dan susut pengeringan. Patch dibuat dalam 3 formula berdasarkan variasi release enhancer berupa gliserin, propilenglikol, dan tween 80 dengan metode solvent casting. Patch yang diperoleh diuji sifat fisika kimia berupa uji keseragaman bobot, uji surface pH, uji swelling index dan uji folding endurance. Uji SIB dilakukan dengan metode alternatif berupa Slug Mucosal Irritation (SMI) berdasarkan presentase mukus yang diproduksi oleh Laevicaulis Alte. Uji SIB dilakukan sebanyak 3 kali dengan waktu kontak 15 menit dan 1 jam istirahat diantara masing-masing waktu kontak. Data presentase total mukus yang diperoleh dianalisis statistik menggunakan SPSS 15 dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa patch ekstrak daun sirih dengan variasi release enhancer dapat menimbulkan sensasi SIB. Patch dengan enhancer gliserin termasuk kategori munculnya sensasi SIB berat, patch dengan enhancer tween 80 termasuk kategori munculnya sensasi SIB sedang dan patch dengan enhancer propilenglikol termasuk kategori munculnya sensasi SIB ringan. Extracts of betel leaf (Piper betle L.) has the main ingredients in the form of eugenol which have antibacterial activity are severe irritant. Extracts formulated in a buccal mucoadhesive patch form for ease of use and patient comfort. Formula which prolonged contact with mucous should be evaluated to predict the emergence of sensation Stinging, Itching, Burning (SIB). This study aims to prove the emergence of sensation SIB buccal mucoadhesive patches of betel leaf extract. Betel leaf extract was made by infundation then freeze drying process afterwards tested specifications include organoleptic test and drying shrinkage. Patch made within 3 formula based on variations in release enhancers such as glycerin, propilenglikol, and tween 80 with a solvent casting method. Patch was tested chemical physical properties such as test weight uniformity, surface pH test, swelling index test and folding endurance test. SIB test performed with an alternative method such as Slug Mucosal Irritation (SMI) is based on the percentage of mucus produced by Laevicaulis Alte. SIB test done 3 times with a contact time of 15 minutes and 1 hour break between each contact time. The percentage of total mucus were analyzed statistically using SPSS 15 with a 95% confidence level. The results showed that the patch betel leaf extract with variations release enhancers can cause a sensation of SIB. Patch with glycerin enhancers including the emergence of sensation SIB severe category, patch with tween 80 enhancer including the emergence of sensation SIB moderate category and patch with propilenglikol enhancer including cthe emergence of sensation SIB mild category.
