EFEK EKSTRAK JAHE MERAH (Zingiber officinale Rocs. var. rubra) TERHADAP DAYA HAMBAT Escherichia coli ISOLAT BABI

Jahe merah telah diketahui mengandung senyawa bioaktif golongan fenol (shagaol dan gingerol), flavonoid, terpenoid dan minyak atsiri, serta memiliki aktivitas sebagai antibakteri, antivirus, dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui efek ekstrak jahe merah terhadap E. coli isolat babi. Escherichia coli diidentifikasi ulang dengan media Eosin Methylen Blue Agar (EMBA) dan uji biokimia serta uji sensitivitas menggunakan antibiotik: penisilin, gentamisin, amoksilin, streptomisin, tetrasiklin, enrofloksasin, florfenikol, flumequine, doksisiklin, dan amikasin. Uji efek antibakteri jahe merah digunakan metode difusi Kirby-Bauer. Biakan bakteri ditanam pada media Mueller Hinton Agar (MHA), disk ekstrak jahe merah dengan konsentrasi 20%, 30%, 40%, 50%, dan 60% diposisikan pada permukaan agar, diinkubasi pada suhu 370C selama 16-18 jam, zona hambat diukur kemudian dianalisa. Hasil rata-rata pengukuran zona hambat pada konsentrasi 20% 6,8 mm, konsentrasi 30% 7 mm, konsentrasi 40% 7,3 mm, konsentrasi 50% 8,5 mm, dan konsentrasi 60% 11 mm. Berdasarkan hasil penelitian ini, dapat disimpulkan bahwa ekstrak etanol jahe merah tidak mampu menghambat pertumbuhan E. coli isolat babi pada konsentrasi 20%, 30%, 40%, dan 50%, namun menghambat lemah pada konsentrasi 60%.

Red ginger has been known containing bioactive substance of phenol group (shagaol and gingerol), flavanoid, terpenoid and essential oil, which has the activity as antibacterial, antiviral and antiinflammatory. This research is aimed to understand the effect of red ginger extract in inhibiton of E. coli isolates from pigs. Escherichia coli was identified with Eosin Methylen Blue Agar (EMBA) medium, biochemical tests, and sensitifity test using the following antibiotics: penicilin, gentamycin, amoxylin, streptomycin, tetracycline, enrofloxacin, florfenicol, flumequine, doxycycline, and amikacin. For understanding the antibacterial properties of red ginger with Kirby-Bauer diffusion method. The bacteria was grown on Mueller Hinton Agar (MHA) medium, and disks containing red ginger extract at 20%, 30%, 40%, 50% and 60% of concentration was positioned on the surface of the agar, and incubated at 370C for 16-18 hours, inhibition zone was measured and analyzed. The mean result of inhibitory zone at 20% of concentration is 6,8 mm, following by 7 mm at 30% of concentration, 7,3 mm at 40% of concentration, 8,5 mm at 50% of concentration, and 11 mm at 60% of concentration. Based on the result, it can be concluded that red ginger ethanol extract could not inhibit E. coli at 20%, 30%, 40%, and 50% of concentration, but mildly inhibit at 60% of concentration.
