PENGARUH VARIASI KADAR HPMC DAN TWEEN 80 TERHADAP SIFAT FISIK DAN STABILITAS FISIK GEL EKSTRAK AIR KERING TEMULAWAK

Ekstrak air kering temulawak (Curcuma xanthorriza Roxb.) memiliki kandungan kurkumin yang telah diketahui memiliki sifat antibakteri, antiinflamasi, hingga antifungi sehingga berpotensi dimanfaatkan sebagai obat luka. Tujuan dari penelitian ini yaitu membuat sediaan gel dari ekstrak air kering temulawak menggunakan berbagai kadar HPMC dan tween 80 dan melihat pengaruh formulasi terhadap sifat fisik dan stabilitas fisik gel. Sediaan gel dibuat dengan menggunakan variasi kadar HPMC dan tween 80 (3,00%: 1,00%; 5,00%: 1,00%; 3,00%: 2,00%; 5,00%; 2,00%). Gel yang diperoleh dievaluasi sifat fisiknya meliputi uji organoleptis, homogenitas, pH, viskositas, daya sebar, dan daya lekat. Data yang diperoleh dianalisis dengan software Design Expert 9 untuk melihat pengaruh variasi kadar HPMC dan tween 80 terhadap sifat fisik gel dan mendapatkan formula optimum. Keempat formula dilakukan uji stabilitas dengan centrifugation test sedangkan formula optimum dilakukan uji stabilitas fisik dipercepat selama 3 bulan. Uji stabilitas dipercepat menggunakan analisis one-way ANOVA untuk melihat ada atau tidaknya perbedaan sifat fisik gel optimum selama penyimpanan 3 bulan. Hasil penelitian menunjukkan bahwa HPMC memberi pengaruh dominan terhadap viskositas, daya sebar, dan daya lekat gel, namun HPMC dan tween 80 tidak memberikan pengaruh terhadap sifat organoleptis, homogenitas, dan pH gel. Formula optimum gel yang diperoleh yaitu dengan kadar HPMC 5,00% dan tween 80 1,00%. Formula a (HPMC: tween 80 = 5,00% : 1,00%) dan formula ab (HPMC: tween 80 = 5,00% : 2,00%) stabil secara fisik setelah disentrifugasi. Formula optimum gel stabil secara organoleptis, homogenitas, pH, viskositas, dan daya lekat, namun daya sebar gel tidak stabil selama penyimpanan 3 bulan.

Water dry extract of temulawak (Curcuma xanthorrhiza Roxb.) contains curcumin which known had antibacterial, antiinflammation, and antifungal activity so that it has potential used as wound healing. The purpose of this study was formulating gel of water dry extract of temulawak made by variation of HPMC and tween 80 concentrations and investigating the influence of formulations on physical characteristics and physical stabilities gel. Gel was made by variation of HPMC and tween 80 concentrations (3,00%: 1,00%; 5,00%: 1,00%; 3,00%: 2,00%; 5,00%; 2,00%). Gel preparations was evaluated the physical characeristics by organoleptic test, homogenity test, pH, viscosity, spreadability, and adhesiveness. The physical properties were analyzed by Design Expert 9 software to investigate the influence of variation of HPMC and tween 80 on physical characteristics gel and to obtain optimum formula. The physical stabilities test of four formulas using centrifugation test whereas optimum formula using accelerated testing during 3 months. Accelerated testing of gel were analyzed by one-way ANOVA to see the differences physical characteristics of optimum formula during 3 months storage. The results showed that HPMC had dominant influenced on viscosity, spreadability, and adhesiveness gel, but HPMC and tween 80 had not infuence on organoleptic, homogenity, and pH gel. Optimum formula were HPMC 5,00% and tween 80 1,00%. Formula a (HPMC: tween 80 = 5,00% : 1,00%) and formula ab (HPMC: tween 80 = 5,00% : 2,00%) were stable after centrifugation. Optimum formula were stable on organoleptic, homogenity, pH, viscosity, and adhesiveness, however spreadability of gel was not stable during 3 months storage.
