PENGARUH PEMBERIAN PARACETAMOL BERBAGAI DOSIS TERHADAP KADAR KREATININ DAN BLOOD UREA NITROGEN PADA TIKUS WISTAR

Peningkatan kadarkreatinin dan BUN merupakan indikator kerusakan ginjal. Kerusakan ginjal salah satunya bisa terjadi karena konsumsi obat-obatan. Paracetamol adalah salah satu jenis obat analgesik dan antipiretik, namun pada dosis berlebihan paracetamol dapat menyebabkan kerusakan pada ginjal sebagai organ ekskresi utama, akibat akumulasi metabolit nefrotoksin NAPQI.Penelitian ini bertujuan untuk melihat pemberian berbagai dosis paracetamol terhadap peningkatan kadar kreatinin dan BUN. Dua belas ekor tikus Wistar betina usia 6 minggu, berat 74,9-81,9 gram yang terbagi dalam 4 kelompok : Kelompok kontrol (K), P1 diberi paracetamoldosis 750 mg /Kg BB, P2 diberi paracetamol dosis 1500 mg/kg BB, dan P3 diberi paracetamoldosis 3000 mg/Kg BB. Kadar kreatinin dan BUN diperoleh dari darah yang diambil pada hari ke-3 setelah pemberian paracetamol dengan lalu dihitung kadar kreatinin dan BUN dengan metode Diasys Creatinin FS dan Dyasys Urea FS. Hasil penelitian memperlihatkan bahwa tidak ada peningkatan bermakna kadar kreatinin dan BUN (p>0,05) pada kelompok yang diberi paracetamol dengan varian dosis di hari ke-3 setelah pemberian paracetamol. Ini menunjukan pemberian paracetamol dosis 750 mg/Kg BB, 1500 mg/Kg BB, dan 3000 mg/Kg BB tidak menyebabkan peningkatan kadar kreatinin dan BUN pada hari ke-3 pasca paparan paracetamol.

The increasing level of creatinin and Blood Urea Nitrogen (BUN) can be used as an indicator of the kidney damage. Kidney damage can occur due to consumption of drugs. Paracetamol is one of analgesic and antipyretic drug. Nevertheless, the use of paracetamol in excessive doses can damage kidney as the main excretory organ due to the accumulation of metabolites nephrotoxin NAPQI of paracetamol. This study aimed to observe the administration of various doses of paracetamol to creatinin and BUN level. Twelve female wistar rats with age of 6 weeks weight 74.9 to 81.9 gram were divided into 4 groups: control group (K), P1were given paracetamol dose of 750 mg / kg bw, P2 were given paracetamol dose of 1500 mg / kg, P3 given paracetamol dose of 3000 mg / kg bw. Creatinin and BUN levels were obtained from blood which were taken at the third days after giving paracetamol. Then, creatinine and BUN levels were calculated using the Diasys Creatinine FS and DiaSys Urea FS method. The results of this research indicated that there were no significant increase in creatinin and BUN level (p>0,05)in groups paracetamol with various doses at the third days after paracetamol administration. This shown that administration of paracetamol dose of 750 mg / kg bw, 1500 mg / kg bw, and 3000 mg / kg bw has no significant increase in creatinin and BUN level in Wistar rat at the third days after paracetamol administration.
