PENGARUH APLIKASI TOPIKAL GEL EKSTRAK KULIT MANGGIS (Garcinia mangostana L.) TERHADAP ANGIOGENESIS TULANG ALVEOLAR PADA PERIODONTITIS Sprague dawley

Periodontitis merupakan keadaan inflamasi pada jaringan periodontal yang disebabkan oleh mikroorganisme yang tidak dilakukan perawatan sehingga inflamasi terus berkembang. Tubuh akan melakukan reaksi saat terjadinya kerusakan yang bertujuan memperbaiki fungsi maupun strukutur jaringan menjadi normal kembali, dengan angiogenesis sebagai salah satu parameternya. Kulit manggis memiliki potensi dalam membantu proses penyembuhan karena kandungan xanton dan saponin. Penelitian ini bertujuan untuk mengetahui pengaruh aplikasi gel ekstrak kulit manggis (Garcinia mangostana L.) terhadap angiogenesis tulang alveolar pada periodontitis Sprague dawley. Tiga puluh enam tikus Sprague dawley jantan berumur 3 bulan, dibagi dalam 3 kelompok (perlakuan, kontrol positif, dan kontrol negatif). Kelompok perlakuan dilakukan ligasi pada gigi incisivus rahang bawah. Kelompok perlakuan diaplikasikan gel ekstrak kulit manggis 10%, kelompok kontrol positif diaplikasikan gel ibuprofen 10%, dan kelompok kontrol negatif diaplikasikan gel CMC-Na 2%, selama 2 minggu masing-masing diaplikasikan 1 kali sehari selama 1 menit secara topikal. Jaringan yang rusak diambil pada hari ke-3, 5, 7, dan 14 lalu dibuat preparat histologis dengan pewarnaan Hematoksilin-Eosin. Hasil uji analisis two-way ANOVA menunjukkan terdapat perbedaan yang signifikan (p<0,05) antara aplikasi, hari pengamatan, serta interaksi aplikasi dengan hari pengamatan terhadap jumlah pembuluh darah baru. Kesimpulan dari penelitian ini adalah aplikasi gel ekstrak kulit manggis 10% meningkatkan jumlah pembuluh darah baru pada periodontitis Sprague dawley.

Periodontitis is an inflammatory condition in periodontal tissue that caused by microorganisms that will keep growing if it doesn't treated directly. Our body have it's own defence mechanism that cause the body to heal that repaired the function and the injured structure to normal condition, with angiogenic as one of the parameter. Manggis peel have the potential in helping the healing process because it contains xanton and saponin. This research goal is to study the impact of manggis skin extract gel (Garcinia mangostana L) application on angionenic alveolar bone with periodontitis on Sprague dawley rat. Thirty six male Sprague dawley rats age of 3 months old, were divided into 3 groups (treatment, positive control, and negative control). Treatment group were ligated on the mandible in incisivus teeth. The application of extract manggis by 10% was done on treatment group, whereas the positive control group got applied by 10% of ibuprofen gel and 2% of CMC-Na got applied on the negative group, 1 time of application for two weeks full was done on each group that lasted for one minute topically. Damaged tissue taken on day 3, 5, 7, and 14 then it converted to histological sample with the stain of Hematoksilin-Eosin. The result of two-way ANOVA analysis showed that there was a significant difference (p<0,05) between application of gel, day of observation, and also the interaction application with the day of observation the number of new blood vessels. Conclusion of this resarch is the application of extract manggis skin gel by 10% increase the number of new blood vessels on Sprague dawley's periodontitis.
