IDENTIFIKASI DAN KARAKTERISASI Staphylococcus aureus ISOLAT ASAL AYAM SERTA UJI PATOGENISITAS BERDASARKAN DAYA FAGOSITOSIS NEUTROFIL

Staphylococcus aureus merupakan bakteri yang umumnya dikaitkan dengan penyakit stafilokokis. Penelitian ini bertujuan untuk identifikasi, karakterisasi, dan uji patogenisitas Staphylococcus aureus isolat asal ayam yang mengalami artritis, bumble foot dan kepala bengkak. Total isolat yang dipakai sebanyak 44 isolat (13 isolat diambil dari ayam yang mengalami artritis; 8 isolat dari ayam yang mengalami bumble foot, 23 isolat dari ayam yang mengalami kepala bengkak). Identifikasi dilakukan melalui uji Manitol Salt Agar (MSA), pewarnaan Gram, uji katalase, uji clumping factor, uji coagulase, dan uji Voges-Proskauer (VP). Karakterisasi bakteri dilakukan melalui uji sifat hemolisa, produksi pigmen, aglutinasi darah kelinci, dan pertumbuhan koloni pada Soft Agar (SA). Uji patogenisitas bakteri dilakukan berdasarkan daya fagositosis neutrofil terhadap Staphylococcus aureus. Berdasarkan hasil identifikasi 44 sampel terdapat 14 isolat positif Staphylococcus aureus berdasarkan hasil positif pada media MSA, uji katalase, clumping factor, koagulase dan VP. Staphylococcus aureus mampu menghasilkan hemolisa α-hemolisa (50%), β-hemolisa (42,8%), dan γ-hemolisa (7,1%) Staphylococcus aureus juga mampu mengaglutinasi eritrosit (100%), pigmen putih, tumbuh difus (50%) dan kompak (50%) pada media Soft Agar. Staphylococcus aureus dengan koloni difus difagosit oleh neutrofil rata-rata sebanyak 14,2 bakteri/sel sedangkan koloni kompak difagosit rata-rata sebanyak 14 bakteri/sel.

Staphylococcus aureus is a bacteria that common related to the staphylococcis case. The purpose of this study were to identify, characterize, and analyse the pathogenicity level of Staphylococcus aureus that was isolated from chickens with arthritis, bumble foot and swollen head cases. Total of 44 isolates were used in the research including 13 isolates from chickens with arthritis, 8 isolates from chicken with bumble foot and 23 isolates from chicken with swollen head disease. Identification of the isolates were conducted based on manitol salt agar assay, Gram staining, catalase, clumping factor, coagulase tests, and Voges-Proskauer test. Characterization of the isolates were coaluated by hemolysis-production test in the Blood Agar Media, pigmen production, and the growth of colonies in the Soft Agar (SA) media. The pathogenicity of Staphylococcus aureus was examinated by neutrophil phagocytosis-based test. The results of research showed that 14 isolates positive for Staphylococcus aureus based on the ability to fermentation manitol in MSA, positive for catalase, coagulase, clumping factor, and VP tests. All isolates of Staphylococcus aureus showed the ability to produce α-hemolisis (50%), β-hemolisis (42,8%) and γ-hemolisa (7,1%). All Staphylococcus aureus isolate could aglutinate erytrosytes, produce white pigment, and growth with diffuse colonies (50%) and compact (50%) in the Soft Agar (SA). The pathogenicity level based on neutrophil-phagocytosis test showed that Staphylococcus aureus with diffuse colonies could be phagocytosis by neutrophils for about 14.2 bacteria/cell, while the compact colonies could be phagocytosis by neutrophils for about 14 bacteria/cell.
