SUB-ACUTE TOXICITY OF CURCUMA HEYNEANA INFUSION IN DDY (DEUTSCHLAND, DENKEN, AND YOKEN) MICE: AN EVALUATION ON LIVER FUNCTION

Latar Belakang Curcuma heyneana adalah tanaman yang sering digunakan sebagai obat tradisional, yang lebih sering disebut dengan Temugiring. Dalam C. heyneana, terdapat beberapa bahan aktif yang juga terdapat pada tanaman C. longa L, seperti curcumin, demethoxycurcumin, dan isocucumenol,yang ditemukan sebagai hepatotoksik pada konsentrasi tertentu. Belum ada studi sebelumnya yang mengenai toksisitas sub-akut C. heyneana pada mencit dengan parameter fungsi hati (ALT, Bilirubin total, dan Protein total) Tujuan Mengetahui apakah C. heyneana bersifat toksik atau tidak, dan menentukan konsentrasi toksik C. heyneana per oral secara sub-akut pada mencit DDY, dengan mengevaluasi fungsi liver. Metode Studi ini menggunakan Non Ekuivalen Studi Kuasi Eksperimental Post test only. Studi ini menggunakan mencit DDY, dengan total sampel 30 mencit, yang dibagi menjadi 6 grup dengan perlakuan konsentrasi infus C. heyneana yang berbeda beda (0, 100, 200, 300, 400, 500 mg/KgBB) selama 28 hari secara oral. Data kemudian di analisis menggunakan ANOVA satu arah. Hasil Hasil untuk tes ALT(control grup, I, II, III, IV, V) )adalah 45,60 ± 8,14; 59,40 ± 7,44 ; 114,00 ± 88,50 ; 68,40 ± 13,13 ; 56,00 ± 23,99; 50,00 ± 11,77 secara berurutan. Hasil untuk tes protein total 7,08 ± 0,53; 7,17 ± 0,68; 6,99 ± 0,43; 7,27 ± 0,48; 7,00 ± 0,32; 6,64 ± 0,32 secara berurutan. Dan hasil untuk bilirubin total 0,56 ± 0,24; 0,71 ± 0,28; 0,42 ± 0,02; 0,38 ± 0,05; 0,40 ± 0,13; 0,29 ± 0,32 secara berurutan. Hasil studi menunjukkan bahwa tidak ada efek yang signifikan pada level ALT dan Protein total (p>0.05) setelah pemberian 28 hari oral C. heyneana infus pada mencit DDY. Tetapi ada efek menurun yang signifikan (p<0.05) pada bilirubin total, walaupun hanya pada perbandingan rerata grup 100 mg/kgBB dengan 500 mg/kgBB. Kesimpulan Konsentrasi tertinggi pada studi ini (500 mg/kgBB) tidak memiliki efek yang signifikan terhadap fungi hati, dan tidak dapat dikatakan sebagai toksik pada rentang konsentrasi coba ini.

Background Curcuma heyneana plant are often used as traditional medicine called by 'Temu giring'. There are some active compounds from C. heyneana which is same with C. longa L. is found to be hepatotoxic in some level, such as curcumin, demethoxycurcumin, and isocurcumenol which are known to exhibit liver toxicity. There is no study regarding toxicity of C. heyneana with parameters of liver function tests (ALT, bilirubin total, and protein total) Objective To determine the sub-acute Toxic concentration of C. heyneana infusion given in DDY mice per oral by evaluating the liver function Methods This study use Non equivalent Quasi Experimental study with post test only design. This study used DDY mice, the total sample is 30 divided into 6 group (Group control, I, II, III, IV, V) with different concentration of C. heyneana infusion (0, 100, 200, 300, 400, 500 mg/KgBB respectively) for 28 days per oral. The data was analyzed using One Way ANOVA. Results The result for mean of ALT for each group (Group control, I, II, III, IV, V) are 45,60 ± 8,14; 59,40 ± 7,44 ; 114,00 ± 88,50 ; 68,40 ± 13,13 ; 56,00 ± 23,99; 50,00 ± 11,77 respectively. Result for mean of protein total are 7,08 ± 0,53; 7,17 ± 0,68; 6,99 ± 0,43; 7,27 ± 0,48; 7,00 ± 0,32; 6,64 ± 0,32 respectively. And result for mean of bilirubin total are 0,56 ± 0,24; 0,71 ± 0,28; 0,42 ± 0,02; 0,38 ± 0,05; 0,40 ± 0,13; 0,29 ± 0,32 respectively; liver function test showed that there were no significant effect in ALT and Protein total level after 28 days administration of C. heyneana. But there is a significant decrease in mean of bilirubin total (p<0.05) in mean between group 100 mg/KgBW and 500 mg/KgBW. Conclusion The highest concentration (500 mg/kgBB) doesn't give significant effect to liver function test result, thus not considered as toxic.
