POTENSI EKSTRAK BUNGA PACAR AIR (Impatiens balsamina L.) DALAM PENGHAMBATAN RESPON IMUN MENCIT (Mus musculus L.) YANG DIINDUKSI AUTOIMUN

Quercetin memiliki berbagai potensi terapeutik pada manusia seperti menghambat proliferasi sel, melindungi oksidasi LDL, mencegah agregasi platelet, antikanker, antiinflamasi, antioksidan, dan bersifat kardioprotektif. Senyawa ini merupakan salah satu jenis flavonoid yang dapat larut dalam air, metanol dan etanol. Bunga pacar air (Impatiens balsamina L.) diketahui mengandung quercetin. Penelitian ini dilakukan untuk mempelajari pengaruh ekstrak akuades dan fraksi metanol bunga pacar air terhadap respon imun mencit yang diinduksi autoimun. Bunga pacar air dimaserasi dengan akuades dan metanol. Selanjutnya, dilakukan fraksinasi cair-cair dengan menggunakan n-heksana untuk ekstrak metanol. Kandungan quercetin pada ekstrak akuades dan fraksi metanol dideteksi dan dikuantifikasi dengan analisis HPLC. Mencit diinduksi autoimun dengan menginjeksi intraplantar adjuvan Freund's sebanyak 0,3 ml dan booster pada hari ke-5. Perlakuan diberikan pada hari ke-6 secara oral dengan dosis 10, 100, dan 1000 mg/BB/hari. Skor artritis dilihat setiap 3 hari sekali. Mencit dikorbankan pada hari ke-27, timus diambil dan ditimbang untuk menentukan indeks timus. Selanjutnya, dilakukan pengujian aktivitas dan kapasitas fagositosis serta produksi Nitrat Oksida (NO). Data yang diperoleh dianalisis dengan ANOVA dan uji DMRT untuk mengetahui letak beda nyata. Hasil penelitian menunjukkan fraksi metanol memiliki kandungan quercetin yang lebih tinggi (0,54 μg/mg) daripada ekstrak akuades (0,05 μg/mg). Pemberian fraksi metanol pada semua dosis dan ekstrak akuades dosis 1000 mg/BB mampu menurunkan skor artritis sampai berskoring 1. Pemberian ekstrak akuades menunjukkan aktivitas dan kapasitas fagositosis makrofag yang kembali menjadi normal, tetapi tidak pada pemberian fraksi metanol. Fraksi metanol maupun ekstrak akuades yang diberikan mampu menurunkan konsentrasi NO menjadi normal kembali. Pemberian fraksi metanol dan ekstrak akuades mampu menurunkan indeks timus mencit. Pemberian fraksi metanol dan ekstrak akuades mampu menghambat perkembangan penyakit autoimun rheumatoid artritis.

This research was conducted to study the effect of aquadest extract and methanol fractions of Impatiens balsamina flowers on the immune response of autoimmune induction in mice. Impatiens balsamina flowers macerated with aquadest and methanol. Furthermore, the liquid-liquid fractionation using n-hexane to methanol extract. The content of quercetin in the extract aquadest and methanol fractions were detected and quantified by HPLC analysis. Autoimmune mice induced by injecting Freund's adjuvant intraplantar 0.3 ml and a booster on day 5. Treatment was given on day 6 orally at a dose of 10, 100, and 1000 mg / bw / day. Score arthritis seen every 3 days. Euthanasia of mice on day 27, the thymus removed and weighed to determine the index of the thymus. Furthermore, testing the activity and capacity of phagocytosis and the production of Nitric Oxide (NO). Data were analyzed with ANOVA and DMRT to locate the real difference. The extract aquadest showed activity and capacity of macrophages phagocytic were back to normal, but not on providing the methanol fraction. Fraction of methanol and aquadest extracts were able to reduce the concentration of NO is given back to normal. Giving fraction of methanol and aquadest extract can lower thymus index of mice. Giving fraction of methanol and aquadest extracts of Impatiens balsamina able to inhibit the development of autoimmune rheumatoid arthritis disease.
