(Homoptera: Delphacidae) Dwi Ayu Anggraini 10/304844/BI/8554 INTISARI Paecilomyces lilacinus sebagai salah satu agensia hayati yang dapat digunakan untuk mengendalikan Nilaparvata lugens Stal. Tujuan penelitian ini adalah untuk mengetahui konsentrasi konidia P. lilacinus yang dapat digunakan untuk mengendalikan N. lugens secara optimal. Isolat P. lilacinus diperoleh dan konidianya diperbanyak di Laboratorium Ekologi dan Fisiologi Mikrobia, LIPI-Cibinong. Pengujian patogenitas P. lilacinus terhadap nimfa N. lugens instar ketiga dan keempat dilakukan di Laboratorium Entomologi, Fakultas Biologi, UGM pada bulan Maret-Agustus 2014. Pengujian dilakukan secara acak lengkap dengan empat tingkat konsentrasi konidia dan setiap tingkatan diulang sebanyak tiga kali menggunakan metode tetes langsung diatas permukaan integumen nimfa. Jumlah kematian nimfa diamati selama 72 jam setelah perlakuan. Data mortalitas yang diperoleh dianalisis secara statistik menggunakan ANAVA dan DMRT. Nilai LC50 dan LC95 diperoleh dengan menggunakan Analisis Probit. Hasil penelitian menunjukkan bahwa P. lilacinus patogen terhadap nimfa N. lugens instar ketiga dan keempat. P. lilacinus lebih patogen terhadap nimfa N. lugens instar ketiga. Nilai LC50 instar ketiga dan keempat berturut-turut sebesar 1,5x107 konidia/ml dan 8,5x107 konidia/ml, sedangkan nilai LC95 berturut-turut sebesar 6,7x109 konidia/ml dan 1,2x1011 konidia/ml. Dari hasil uji patogenitas, dapat disimpulkan bahwa P. lilacinus berpotensi sebagai agensia pengendali hayati.
Pathogenicity Paecilomyces lilacinus to Nymphs Nilaparvata lugens Stal. (Homoptera: Delphacidae) Dwi Ayu Anggraini 10/304844/BI/8554 ABSTRACT Ones of the biological control for decrease Nilaparvata lugens Stal. damage have been controlled by Paecilomyces lilacinus. The aim of this research was to know the concetration of P. lilacinus conidia that can be used to control N. lugens optimally. P. lilacinus isolate obtained and mass multiplication of conidia was cultured in Laboratory of Ecology and Microbial Phisiology, LIPI-Cibinong. The used of third and fourth instar nymphs of N. lugens for pathogenicity tests of P. lilacinus was conducted in the Entomology Laboratory, Faculty of Biology, Universitas Gadjah Mada in March to August 2014. The research method used was Complete Random Design with four concentration series and three replications. The application of P. lilacinus conidia infection use direct drop methode on surface of the nymph integumen. The mortality of tested nymphs was observed for 72 hours after treatment. The mortality data obtained was analyzed statistically by using ANOVA and DMRT. The lethal concetration of 50% death nymph (LC50) and 95% death nymph (LC95) were analyzed by using Probit analysis. The results showed that P. lilacinus pathogenic to the third and fourth instar nymphs of N. lugens. The P. lilacinus more pathogenic to the third instar nymph than fourth instar nymph of N. lugens. The value of LC50 of the third and fourth instar nymph of N. lugens respectively were 1,5x107 conidia/ml and 8,5x107 conidia/ml, while the value of LC95 respectively were 6,7x109 conidia/ml and 1,2x1011 conidia/ml. From the results of the pathogenicity tests, it can be concluded that P. lilacinus have potential to be a biological control agent Key words: pathogenicity, Paecilomyces lilacinus, Nilaparvata lugens
