EFEK FRAKSI POLISAKARIDA BUAH MENGKUDU (Morinda citrifolia L.) SEBAGAI KO-KEMOTERAPI DOXORUBISIN TERHADAP PROLIFERASI SEL LIMFOSIT TCD8+ TIKUS WISTAR IN VIVO

2014 | Skripsi | FARMASIDoxorubisin sebagai obat antikanker telah dilaporkan memiliki efek samping dapat menurunkan proliferasi limfosit dan rasio sel limfosit TCD4+/CD8+. Kandungan polisakarida dari buah mengkudu telah diketahui memiliki aktivitas imunostimulator. Tujuan dari penelitian ini adalah mengetahui efek ko-kemoterapi fraksi polisakarida buah mengkudu terhadap penggunaan doxorubisin dan melakukan analisis kadar fenolik total dan susut pengeringan terhadap fraksi polisakarida buah mengkudu. Fraksi polisakarida buah mengkudu diendapkan dengan menggunakan etanol 95% dan diuapkan pada penangas air 37 derajat celcius. Sisanya dikeringkan dengan freeze drying. Uji aktivitas imunostimulator dilakukan dengan melihat pertumbuhan proliferasi sel limfosit TCD8+ tikus Wistar. Sel limfosit darah diisolasi dari vena retro-orbitalis mata, diinkubasi dengan monoclonal antibody PE Rat Anti-Mouse CD3 dan FITC Rat Anti-Mouse CD8 dan dicuci dengan FACS lysing solution dibaca menggunakan BD FACS flowsitometer. Hasil yang diperoleh kemudian dianalisis statistik dengan menggunakan software SPSS versi 19 dengan taraf kepercayaan 95%. Uji kandungan fraksi polisakarida dengan uji kandungan total fenolik menggunakan reagen Folin Ciocalteu dibaca pada spekrofotometer dengan panjang gelombang 765 nm. Dilanjutkan dengan uji susut pengeringan pada suhu 105 derajat celcius selama 30 menit hingga tercapai bobot tetap. Hasil penelitian menunjukkan, fraksi polisakarida buah mengkudu pada dosis 25 mg/kgBB dan 50 mg/kgBB dapat meningkatkan persentase proliferasi TCD8+ masing-masing sebesar 148,04 ± 53,5% dan 55,8 ± 11,49%. Pada dosis 100 mg/kgBB dan 200 mg/kgBB menurunkan persentase proliferasi limfosit TCD8+ masing-masing sebesar 23,89 ± 28,46% dan 25,23 ± 5,57% dibandingkan kelompok kontrol normal tanpa perlakuan. Fraksi polisakarida mengandung kadar fenolik total sebesar 0,012 % dan susut pengeringan sebesar 84,38%.
As an anticancer drug doxorubicin has been reported to have side effects can decrease lymphocyte proliferation and lymphocyte cell ratio TCD4 + / CD8 +. The content of polysaccharides from noni fruit has been known to have immunostimulatory activity. The purpose of this study was to determine the effect of chemotherapy co-polysaccharide fraction of noni to the use of doxorubicin and analyzing total phenolic content and drying shrinkage of the polysaccharide fraction of noni. Noni polysaccharide fraction was precipitated using 95% ethanol and evaporated on a water bath 37 degrees of celcius. The rest is dried by freeze drying. Immunostimulatory activity test is done by looking at the growth of cell proliferation TCD8 + lymphocytes Wistar rats. Cells were isolated from venous blood lymphocytes retro-orbital eye, incubated with monoclonal antibody PE Rat Anti-Mouse CD3 and FITC Anti-Mouse Rat CD8 and washed with FACS lysing solution is read using a BD FACS flowsitometer. The results obtained were then statistically analyzed using SPSS software version 19 with a 95% confidence level. Test content of the polysaccharide fraction with the total phenolic content of the test using the Folin-Ciocalteu reagent spekrofotometer read at 765 nm with a wavelength. Followed by drying shrinkage test at a temperature of 105 degrees of celcius for 30 minutes until a fixed weight. The results showed that the polysaccharide fraction Noni at doses of 25 mg / kg and 50 mg / kg body weight can increase the percentage of proliferation TCD8 + respectively by 148.04 ± 53.5% and 55.8 ± 11.49%. At a dose of 100 mg / kg and 200 mg / kg reduced the percentage of lymphocyte proliferation TCD8 + respectively by 23.89 ± 28.46% and 25.23 ± 5.57% compared to the normal control group without treatment.
