PENGEMBANGAN FILM SENSOR KREATININ BERDASARKAN METODE JAFFE DENGAN MENGIMOBILISASI TOMA-PIKRAT DALAM MATRIKS KITOSAN-PEKTIN

Telah dilakukan sintesis film dengan mengimobilisasi TOMA-pikrat ke dalam matriks kitosan-pektin berdasarkan metode Jaffe. Penelitian ini bertujuan sebagai langkah awal pengembangan sensor kreatinin berbasis film. Film disintesis dengan cara menambahkan pikrat ke dalam larutan kitosan pektin (70:30 w/w), dilanjutkan dengan penambahan TOMA-Cl (Trioctylmethylammonium Chloride) disertai pengadukan sampai campuran homogen. Campuran kitosan-pektin-TOMA-pikrat kemudian dicetak dan dikeringkan pada temperatur 100 °C. Film yang dihasilkan kemudian ditentukan panjang gelombang dan waktu optimumnya. Penentuan standar visual dibuat dengan merendam film ke dalam kreatinin yang konsentrasinya bervariasi pada suasana basa. Aktivitas film diuji pada sampel urin. Hasil karakterisasi spektra IR menunjukkan film kitosan-pektin-TOMApikrat telah berhasil disintesis. Hasil karakterisasi SEM menunjukkan dengan jelas tingkat homogenitas film kitosan-pektin-TOMA-pikrat yang disintesis. Film hasil sintesis dapat digunakan untuk menentukan konsentrasi kreatinin urin dengan rentang konsentrasi 200- 3000 ppm. Hasil uji aktifitas film terhadap kreatinin menunjukkan hasil yang cukup baik (akurasi 17-112% terhadap laboratorium A dan 50-70% terhadap laboratorium B) ketika pengujian dilakukan selama 150 menit dengan perbandingan hasil uji laboratorium.

Creatinine sensing film was synthesized by immobilizing TOMA-picrate into chitosan-pectin matrices using Jaffe method principle. The aim of the research is to study initial step of the developing film-based creatinine sensing. The film was obtained by mixing picrate into chitosan- pectin (70:30 w/w) solution followed by adding a number of TOMA-Cl (Trioctylmethylammonium Chloride) under stiring until homogenous mixture was obtained. The resulted film was templated and dried at 100°C. Resulted film was immersed in creatinin solution under alkaline condition. It was followed by determining of the optimum wavelength and time. Standard visual of creatinine -film was made by immersing the film into standard solution of creatinin under various concentration. The activity test of resulted film was tested at urine sample. IR spectra of resulted film showed that the film of chitosan-pectin-TOMApicrate was successfully produced. The SEM image showed the homogenity level of chitosan-pectin-TOMA-picrate film. The synthesized film can be used to determine urine concentration of creatinine in the range of 200- 3000 ppm The result of activity test compared to standard laboratory test showed good agreement ( 17-112% accuracy relative to laboratory A and 50-70% accuracy relative to laboratory B) when the test was done for 150 minutes.
