EFEK HIPOGLIKEMIK KOMBINASI EKSTRAK LARUT ETANOL HERBA SAMBILOTO (Andrographis paniculata (Burm.f.) Ness) DAN DAUN SAMBUNG NYAWA (Gynura procumbens (Lour.) Merr) TERSTANDAR ANDROGRAFOLID DAN KUERSETIN PADA TIKUS DIABETES DIINDUKSI ALOKSAN

Exploration of medicinal plant combination could be an alternative way for developing medicinal plants in order to provide better result or benefits in disease therapy. Andrographis paniculata (Burm. f.) Ness and Gynura procumbens (Lour.) Merr are two medicinal plants that already well known for those hypoglycemic activity. Hopefully, the combination of both extracts could exhibit better effect than those single extract administration. The aim of this study was to evaluate the hypoglycemic effect of A. paniculata ethanolic extract (APEE) and G. procumbens ethanolic extract (GPEE) combination in alloxan induced hyperglycemic rats. The powder of A. paniculata herbs and G. procumbens leaves were extracted by maceration using ethanol 70% respectively. Phytochemical analysis of andrographolide contained in APEE and quercetin contained in GPEE were performed by TLC-densitometry using stationery phase silica gel F254 and mobile phase chloroform-ethyl acetic-methanol-acetic acid (7:2:0.5:0.5 v/v/v). Hyperglycemic condition in rats was induced with a single dose of 150 mg/kg BW alloxan intraperitoneally. The rats were orally administrated with several kind of dosage APEE:GPEE (300:37.5; 200:75; 100:112.5 mg/kg BW) combination of APEE and GPEE for 15 consecutive days. Hypoglycemiceffect was evaluated by measurement of preprandial and postprandial blood glucose levels and other parameters such as pancreatic islet morphology and pancreatic insulin expression. Andrographolide content in APEE was 17.56% while quercetin content in GPEE was 1.88%. The combination of APEE and GPEE exhibit a better hypoglycemic effect, improvements in pacreatic islet morphology and increased insulin pancreatic expression than that of single treatment of APEE or GPEE. Combination (APEE:GPEE) 100:112.5 mg/kg BW showed better hypoglycemic effect than other combinations. Based on this study, that combination could potentially be developed as a hypoglycemic agent.
