UJI KETERATOGENIKAN EKSTRAK SAMPEL DLBS5447 ® (Zingiber majus) PADA TIKUS BUNTING SPRAGUE DAWLEY TINJAUAN TERHADAP SISTEM SKELETAL JANIN

2014 | Skripsi | FARMASIUji keteratogenikan ekstrak sampel DLBS 5447® (Zingiber majus) pada tikus betina bunting galur Sprague Dawley tinjauan terhadap sistem skeletal janin bertujuan untuk mengevaluasi keamanan ekstrak sampel DLBS5447® (Zingiber majus) bila digunakan pada wanita hamil terutama selama masa organogenesis. Penelitian dilakukan menggunakan tikus betina perawan galur Sprague Dawley dewasa, sehat dengan berat antara 175-250 gram yang memiliki siklus estrus teratur. Hewan uji dibagi menjadi 4 kelompok, yaitu kelompok I (Kontrol) hanya di beri CMC-Na, kelompok II, III, dan IV adalah kelompok perlakuan yang masing-masing diberi ekstrak Sampel DLBS5447® dengan dosis sebesar 37,8 mg/kg BB, 75,6 mg/kg BB, 151,2 mg/kg BB. Tikus dipejani secara oral sekali sehari mulai hari ke-1 sampai hari ke-17 kebuntingan, lalu pada hari ke 20 dilakukan bedah sesar. Janin dipisahkan dari induk lalu diamati sistem skeletal janin kemudian dianalisis dengan uji statistika taraf kepercayaan 95%. Hasil penelitian menunjukkan pada kelompok dosis 2 (75,6mg/kg BB) menyebabkan kelainan rudimentary pada vertebra, sedangkan pada kelompok dosis 3 (151,2mg/kg BB) menyebabkan kelainan partially ossified pada sternebra serta kenaikan kejadian not ossified pada karpal.
Teratogenic test of extract sample DLBS5447® (Zingiber majus) on female pregnant rats Sprague Dawley strain review of the fetal skeletal system aims to evaluate the safety extract sample DLBS5447® (Zingiber majus) when it was used on pregnant women, especially during the period of organogenesis. The research was conducted using the rats Sprague Dawley strain of adult virgin female, healthy, weight between 175-250 grams that have estrus cycle regularly. The rats were divided into 4 groups: Group I (control) was given CMC-Na, Groups II, III, and IV were the treatment groups given extract sample DLBS5447® at a dose of 37.8 mg/kg of body weight, 75.6 mg/kg of body weight, 151.2 mg/kg of body weight. The rats were treated orally once a day from the 1st until the 17th of pregnancy. Then on the 20th day of pregnancy, it was performed a caesarean surgery. The fetus was separated from the mother and then observed the fetal skeletal system then analyzed statistical with confidence level of 95%. The results showed that in the group 2 doses (75,6 mg/kg BW) caused rudimentary abnormalities on the vertebrae, while in the group 3 doses (151,2 mg/kg BW) caused partially ossified abnormalities on the sternebra and increased the incidence of carpal not ossified.
