ISOLASI DAN UJI AKTIVITAS ANTIBAKTERI METABOLIT SEKUNDER DAUN TENDANI (Goniothalamus macrophyllus Hook.f. & Thoms)

2014 | Tesis | S2 Ilmu KimiaIsolasi dan uji aktivitas antibakteri metabolit sekunder daun tendani (Goniothalamus macrophyllus Hook.f. & Thoms) telah dilakukan. Daun tendani diekstraksi dengan metanol dan difraksinasi secara berurutan dengan n-heksana dan etil asetat. Fraksi etil asetat dianalisis dengan GC-MS dan dipisahkan dengan kromatografi kolom menghasilkan 7 fraksi. Fraksi 4 dipisahkan kembali dengan kolom kromatografi menghasilkan fraksi 4.3. Hasil isolasi diidentifikasi menggunakan Spektroskopi Massa dan NMR. Hasil analisis GC-MS ekstrak etil asetat daun tendani menunjukkan kandungan senyawa benzaldehida, spathulenol, metil heksadekanoat, 1,2-dimetoksi-1-fenil etana, phitol, dan stigmasterol. Fraksi 4.3 hasil berupa kristal jarum bening dan diidentifikasi sebagai 8-asetat-7-fenil-2,6-dioksabisiklo[3.3.1]nonana-3-on atau 9-deoksigoniopypyron asetat. Diameter zona hambat pertumbuhan bakteri Staphylococcus aureus ekstrak metanol, nheksana, dan etil asetat dengan konsentrasi 5% berturut-turut sebesar 7,40, 2,50 dan 6,18 mm, sedangkan diameter zona hambat pertumbuhan bakteri Escherichia coli adalah 6,63, 2,46 dan 4,07 mm. Diameter zona hambat senyawa 9-deoksigoniopypyron asetat terhadap Staphylococcus aureus dan Escherichia coli pada konsentrasi 0,2% masing-masing 6,27 mm dan 5,25 mm
Isolation and evaluation of antibacterial activities of secondary metabolites of tendani leaves (Goniothalamus macrophyllus Hook.f. & Thoms) have been conducted. Tendani leaves were extracted with methanol and fractionated sequentially with n-hexane and ethyl acetate. The ethyl acetate extracts were analyzed using GC-MS and separated by column chromatography to afford 7 fractions. Fractions 4 were rechromatographed to give fraction 4.3. The structure of isolated compound was identified by Mass Spectroscopy and NMR. The GC-MS analysis shows ethyl acetate extracts of tendani leaves contain benzyldehyde compounds, sphatulenol, methyl hexadecanoat, 1,2-dimethoxy-1-phenyl-ethane, phytol, and stigmasterol. Fraction 4.3 is colorless needle-shape crystals and identified as 8-acetate-7-phenyl-2,6-dioxabicyclo[3.3.1]nonane-3-one or 9-deoxygoniopypyrone acetate. Diameters of inhibition zone of methanol extracts, n-hexane, and ethyl acetate with concentration of 5% against Staphylococcus aureus bacteria are 7.40, 2.50 and 6.18 mm respectively, whereas against Escherichia coli are 6.63, 2.46 and 4.07 mm respectively. Diameters of inhibition zone of 9-deoxygoniopypyrone acetate with concentration of 0.2% against Staphylococcus aureus and Escherichia coli are 6.27 mm and 5.25 mm respectively
