PEMBUATAN FILM KITOSAN-PEKTIN DENGAN PENAMBAHAN EMULSIFIER ASAM STEARAT SEBAGAI MATRIKS PELEPASAN KURKUMIN

Telah dilakukan penelitian tentang studi pelepasan kurkumin menggunakan film kitosan-pektin dengan penambahan emulsifier asam stearat. Penelitian ini bertujuan untuk mengetahui efektivitas film dalam pelepasan kurkumin di dalam larutan buffer:etanol (v/v). Di dalam penelitian ini telah dipelajari pengaruh komposisi kurkumin, komposisi asam stearat, komposisi etanol dan variasi pH terhadap laju pelepasan kurkumin. Film kitosan-pektin dibuat dengan cara menambahkan sejumlah kitosan dalam larutan asam asetat 1% ke dalam larutan pektin, kemudian ditambahkan asam stearat dalam isopropanol. Film yang dihasilkan dikarakterisasi dengan FT-IR. Studi pelepasan kurkumin dilakukan dengan cara film direndam ke dalam larutan buffer-etanol (v/v) selama 6 jam, kemudian dianalisis menggunakan spektrofotometer UV-Vis. Kinetika pelepasan kurkumin dianalisis menggunakan persamaan Korsmeyer-Peppas. Hasil penelitian menunjukkan bahwa pelepasan kurkumin dipengaruhi oleh komposisi kurkumin yang termuat, komposisi asam stearat pada film dan pH larutan buffer. Laju pelepasan optimum dari film dengan ketebalan 0,11 mm pada larutan buffer pH 7 dalam 40% etanol diperoleh ketika jumlah kurkumin yang termuat dan asam stearat masing-masing adalah 54,5 dan 45,4 mg/g film, sedangkan kurkumin yang terlepas sebesar 66,04%. Kinetika pelepasan kurkumin mengikuti model Korsmeyer-Peppas dengan konstanta laju (k) sebesar 7,53 menit-0,38 dan n sebesar 0,38. Mekanisme pelepasan kurkumin didominasi oleh faktor difusi.

The study was designed to prepare curcumin releasing matrix using kitosan-pectin films with the addition of stearic acid as an emulsifier. The aim of this research was to identify the effect of variation of the pH of the buffer solutions, the amount of loaded curcumin, the amount of stearic acids in the films toward the rate and the effectiveness of the films to release curcumin. The chitosan-pectin films was prepared by adding certain amount of chitosan in 1% acetic acid solution into 1% aqueous pectin solution, then isopropanol solution of stearic acid was added. The films obtained were characterized using FT-IR spectrophotometer. The study of curcumin release was performed by soaking film in buffer-ethanol solutions for 6 hours, and the released curcumin was measured by UV-Vis spectrophotometer. The kinetics of releasing curcumin was analyzed by Korsmeyer-Peppas equations . The result of this study showed that the releasing of curcumin was influenced by the amount of loaded curcumin, the amount of stearic acid in the film and buffer pH of the solution. The optimum releasing rate from the film thickness of 0.11 mm at buffer pH 7 in 30% ethanol solution was obtained when the amount of curcumin loaded and stearic acid were 54.5 and 45.4 mg/g film, respectively, which gave the curcumin release of 66.04%. The kinetics of the curcumin release followed a Korsmeyer-Peppas model with rate constant (k) of 7.53 minute-0.38 and n of 0.38. The curcumin release mechanism was mainly dominated by diffusion mechanism
