ASSOCIATION BETWEEN ENDOTHELIAL NITRIC OXIDE SYNTHASE (ENOS)EXPRESSION AND VASCULAR REMODELING IN ACUTE KIDNEY INJURY MODEL AFTER ISCHEMIC/REPERFUSION EPISODE IN MICE

BACKGROUND: Acute Kidney Injury is well known for its clinical problem with Ischemic/Reperfusion(I/R) injury as the most common cause. eNOS is assumed to has protective effect to the renal vascular. AKI can be detected by serum creatinine level measurement and vascular remodeling observation. Therefore, the association between eNOS and vascular remodeling was made. OBJECTIVES: To elucidate the association between eNOS and vascular remodeling in acute kidney injury after ischemic-reperfusion episode in mice. METHODS: Twenty four Balb-C mice were divided into 4 groups; SO(n=7), 1Hr I/R(n=5), 3Hr I/R(n=5), and 24Hr I/R (n=7). Bilateral renal pedicles clamping for 30 minutes was performed in 1Hr I/R, 3Hr I/R, and 24Hr I/R, followed by reperfusion for 1 hour, 3 hours, and 1 day, respectively. The blood was taken from scleral sinus to measure creatinine serum level. eNOS expression was examined by using RT-PCR. The kidneys were taken and vascular remodeling was observed. RESULTS: In this study, The manifestation of AKI was proved by significant increased serum creatinine level comparison between SO, 1Hr I/R, 3Hr I/R, and 24Hr I/R (p < .001) (p < .005). Reduction of eNOS expression was showed significant result in 24Hr I/R as compared to SO group(p = .028)(p < .005). Vascular remodeling was observed by the presence of significant increased of arterial wall thickness (p = .001) (p < .005) and significant decrease of lumen to wall area ratio(p = .003) (p < .005) in 24Hr I/R as compared to SO group. CONCLUSION: There is a negative association between eNOS expression reduction and vascular remodeling.
