CORRELATION BETWEEN ENDOTHELIAL NITRIC OXIDE SYNTHASE (ENOS) EXPRESSION AND TUBULAR EPITHELIAL INJURY IN ACUTE KIDNEY INJURY MODEL AFTER ISCHEMIC-REPERFUSION INJURY IN MICE

BACKGROUND: Acute Kidney Injury (AKI) is still one of the most common and serious clinical problem. Examination on the level of nitrogenous wastes shares their own limitations. Therefore, endothelial Nitric Oxide Synthase (eNOS) expression which is suspected to have a renal protective effect against the ischemicreperfusion injury induced AKI, was further studied. OBJECTIVES: To identify the correlation between eNOS expression and tubular epithelial injury in mice suffering from AKI after ischemic-reperfusion injury. METHODS: Twenty four mice with Balb C background were divided into Sham Operation (SO) (n=7), 1 hr I/R (n=5), 3 hr I/R (n=5), and 24 hr I/R (n=7) groups. In 1 hr I/R, 3 hr I/R, and 24 hr I/R groups, bilateral clamping of renal pedicles were done for 30 minutes, followed by reperfusion for 1 hour, 3 hours, and 24 hours, respectively, but not in SO group. Tubular epithelial injury score, serum creatinine level, and eNOS expression were examined in this study. RESULTS: Significant increments of tubular epithelial injury grade and serum creatinine level from SO, 1 hr I/R, 3 hr I/R, and 24 hr I/R groups were observed (p=.002 and p<.001, respectively). The eNOS expression decreased significantly in 24 hr I/R group compared to SO group (p=.028). The decreased eNOS expression and increased tubular epithelial injury score were highly correlated even though the result was non-significant statistically (r=-.866, p =.167). CONCLUSION: The expression of eNOS demonstrated a tendency of inverse correlation with the tubular epithelial injury.
