PENGARUH PEMBERIAN EKSTRAK ETER DAUN OCIMUM BASILICUM TERHADAP INDEKS ARTHRITIS TIKUS PUTIH YANG DIINDUKSI DENGAN COMPLETE FREUND’S ADJUVANT

Latar Belakang : Ocimum basilicum atau kemangi merupakan tanaman dengan banyak manfaat, mudah ditemukan dan sering digunakan oleh masyarakat Indonesia. Manfaatnya sebagai anti inflamasi telah terbukti pada inflamasi akut. Sampai sekarang, belum pernah dilakukan penelitian mengenai pengaruh Ocimum basilicum terhadap inflamasi kronis. Salah satu bentuk inflamasi kronis adalah rheumatoid arthritis. Tujuan : Mengetahui pengaruh pemberian ekstrak eter daun Ocimum basilicum terhadap indeks arthritis tikus putih yang diinduksi dengan Complete Freund’s Adjuvant (CFA). Metode : Tikus wistar sebanyak 40 ekor dikelompokkan ke dalam 5 kelompok perlakuan sama banyak. Pada minggu pertama dan ke-4 tikus diinduksi dengan CFA masingmasing sebanyak 0,05 ml. Pada minggu ke-6 hingga minggu ke-9 setiap tikus mendapat intervensi per-oral setiap hari. Kelompok I, II dan III mendapat ekstrak Ocimum basilicum dengan dosis 50 mg/2 ml, 100 mg/2 ml, dan 200 mg/2 ml. Sebagai kontrol, kelompok IV mendapat Na diklofenak 2 mg/2 ml dan kelompok V mendapat aquades 2 ml. Indeks arthritis tikus dievaluasi menggunakan skala skor setiap minggu. Delta indeks arthritis sebelum dan sesudah perlakuan dianalisa dengan uji non parametrik Kruskal-wallis dan dibandingkan antar kelompok dengan uji non parametrik Mann-whitney. Hasil dinyatakan bermakna jika p<0.05 dan dinyatakan tidak bermakna jika p>0.05. Hasil : Tidak ada perbedaan efek yang bermakna antar variasi dosis (p>0.05). Rata-rata penurunan indeks arthritis oleh dosis I 0,91 ± 0,23 (p=0,011), dosis II 1,03 ± 0,36 (p=0,007), dan dosis III 0,97 ± 0,25 (p=0,007) signifikan lebih besar dibandingkan kontrol negatif 0,50 ± 0,27. Rata-rata penurunan indeks arthritis oleh dosis I (p=0,003), dosis II (p=0,031), dan dosis III (p=0,007) signifikan lebih kecil dibandingkan kontrol positif 1,38 ± 0,23. Kesimpulan : Pemberian ekstrak eter daun Ocimum basilicum dengan dosis 50 mg/2 ml, 100 mg/2 ml, dan 200 mg/2 ml dapat menurunkan indeks arthritis tikus putih yang diinduksi Complete Freund’s Adjuvant (CFA).

Background : Ocimum basilicum or sweet basil is a plant with many benefits, easy to find and often used by Indonesian society. Its anti-inflammatory properties has been shown in acute inflammation. Thus far, the research on the effect of Ocimum basilicum against chronic inflammation has not yet conducted. One form of chronic inflammation is rheumatoid arthritis. Objective : To find out the effect of Ocimum basilicum leaves ether extract administration towards white rats’ arthritis index that has been induced by Complete Freund’s Adjuvant (CFA). Methods : This study used 40 white wistar rats divided into 5 treatment groups equally. On the first and fourth week, the rats induced using CFA 0,05 ml. On the sixth until ninth week, the rats were given oral intervention daily. First, second, and third group get the Ocimum basilicum extract in dose range of 50 mg/ 2ml, 100 mg/2 ml, and 200 mg/2 ml. As control, fourth group get Na diclofenac 2 mg/2 ml and fifth group get 2 ml of Aquades. The arthritis index was evaluated every week. Arthritis index delta from before and after treatment was analyzed using Kruskal-wallis non parametric test and compared between groups using Mannwhitney non parametric test. Results revealed significant if p<0.05 and not significant if p>0.05. Results : There is no significant differences effect among three dose variance (p>0,05). Arthritis index average decline shown by dose I 0,91 ± 0,23 (p=0,011), dose II 1,03 ± 0,36 (p=0,007), and dose III 0,97 ± 0,25 (p=0,007) are significantly bigger than negative control group 0,50 ± 0,27. Arthritis index average decline shown by both dose I (p=0,003), dose II (p=0,031), and dose III (p=0,007) are significantly smaller than the positive control group 1,38 ± 0,23. Conclusion : Administration of 50 mg/2 ml, 100 mg/2 ml, and 200 mg/2 ml Ocimum basilicum leaves ether extract showed a decrease on white rats’ arthritis index induced by Complete Freund’s Adjuvant (CFA).
