Pengaruh kurkumin analog (Pentagamavunon-O) pada produksi progesteron oleh kultur sel luteal tikus

Pentagamawnon-0 merupakan senyawa sintetis analog kurkumin yang mempunyai potensi sebagai obat pengatur kesuburan dan belum diketahui mekanisme kerjanya pada sistem reproduksi. Penelitian ini bertuj uan untuk mengkaji pengaruh pentagamavunon-0 pada kultur sel Iuteal tikus dengan mengukur kadar produksi progesteron yang sebeluinnya diberi kan perangsangan LH, PGF?,, dan teofilin. Korpus luteum tikus umur 4 hari dikultur dalam media MEM yang inengandung fetal bovine serum 10 %. Kultur sel luteal dikelompokkan inenjadi I6 kelompok dalam 10 ulangan (n=10). Kelompok 1-8 tanpa mendapat perlakuan teofilin sebagai keloinpok A dan keloinpok 9-1 6 inendapat perlakuan teofilin sebagai kelompok B. Masing-masing kelompok mendapat perlakuan : Pelarut (Gl), PGV-0 (G2), LH (G3), LH+PGV-0 (G4), PGF2, (G5), PGF:, + PGV-0 (G6), LH+PGF?, (G7), LH +PGF2, + PGV-0 ((38). Seluruh perlakuan diinkubasi selama 24 jam dan selanjutnya kadar progesteron yang terdapat di dalam media diukur menggunakan metode Radio Immuno Assay. Data yang diperoleh dianalisis secara statistik dengan Analisis Varians satu jalan dilanj utkan uji-t dan menunjukkan perbedaan bermakna pada p0,05). Dari hasil penelitian ini &pat diambil kesiinpulan bahwa : 1). Produksi progesteron oleh KSL pada kontrol dengan pemberian pelarut tidak diturunkan oleh PGV-0, 2). Peningkatan produksi progesteron akibat perangsangan LH diturunkan oleh PGV-O,3). Peningkatan produksi progesteron oleh KSL pa& akumulasi CAMP akibat perangsangan teofilin tidak diturunkan oleh PGV-0.

Pentagamavunon-0 is a synthetic agent analog curcumin that is a potensial drug for fertility regulation.The activity and mechanism in the reproductive system has not known. The objective of this experiment was to study the effect of curcumin analog pentagamavunon-0 on progesterone production by cultured rat luteal cells that stimulated by LH, PGF2, and theophylline. The subject in this study was 4 days old corpus luteum of rat which cultured in MEM medium containing 10 % fetal bovine serum. The homogenous luteal cell cultures were randomly divided into 16 group of treatments in ten replicates (n=10). Group 1 to 8 in the absence of theophylline as group A and group 9-16 in the presence of theophylline as group B. Each of which was administered : solvent (GI), PGV-0 (G2), LH (G3), LH+PGV-0 (G4), PGF:, (G5), PGF,,+PGV-0 (G6), LH+PGF2, (G7) and LH+PGF:,+PGV-0 (GS). All these treatments were incubated for 24 hours and the progesterone concentration in culture media were measured using Radio Immuno Assay (RIA) method. The data collected then were analyzed with one way Analysis of Variance. Differences between means were evaluated by t-test and p<0,05 was taken as level of significance The result of this study showed that the progesterone concentration (nM/lO' cells!m1/24 h) produced by luteal cell cultures in goup A (group without theophylline) is as follow : The level of progesterone produced after treatment by solvent/ control progesterone is 3,94 k I ,65. Progesterone concentration after stimulation by LH is 6,44 k 1,73. It was enhanced significantly compared to control progesterone, The level of progesterone production by coincubation of luteal cell cultures with LH + PGV-0 is 3,46 k 0,76 and LH+ PGF*,+ PGV-0 is 3,93 k 0,68. These results showed that both PGV-0 and PGV-0 + PGF2, were decreased significantly (P<0,05) compared to progesterone production stimulated by LH. These results in group B ( group with theophylline) showed that : The level of progesterone produced by theophylline alone is 5,94 k 1,08. The level of progesterone produced after treatment by PGV-0 is 6,43 f 2,20 and LH+PGV-0 is 6,37 k 1,07. These level of progesterone productions were not significantly different (P>0,05). Based on the result of the research, it is concluded that : 1). The level of control progesterone produced by cultured rat luteal cells was not reduced by PGV-0, 2). Enhancement of progesterone production stimulated by LH was reduced by PGV-0, 3). Enhancement of progesterone production stimulated by theophylline was not reduced by PGV-0.
