STUDI PELEPASAN IN VITRO KURKUMIN DARI FILM KALSIUM ALGINAT DENGAN PENAMBAHAN EMULSIFIER TWEEN-80 DAN SPAN-80

Telah dilakukan penelitian tentang laju pelepasan kurkumin dari film kalsium alginat (Ca-Alg) dengan penambahan emulsifier Tween-80 dan Span-80. Pengaruh sifat hidrofilik-hidrofobik pengemulsi makanan dikaji pada proses pelepasan kurkumin dari film Ca-Alg. Film alginat teremban kurkumin yang mengandung emulsifier Tween-80 dan Span-80 bervariasi dibuat dengan menggunakan larutan kalsium klorida (CaCl2) sebagai agen penaut silang. Dalam penelitian ini, digunakan campuran emulsifier Tween-80 dan Span-80 yang memiliki nilai Hydrophile-Lipophile Balance (HLB) antara 11,9 hingga 14,0. Film yang diperoleh dikarakterisasi dengan Spektrometer IR dan SEM (Scanning Electron Microscopy). Laju pelepasan kurkumin dari film Ca-Alg diteliti secara in vitro selama 8 jam. Konsentrasi kurkumin pada berbagai rentang waktu ditentukan secara Spektrofotometri UV-Vis. Karakterisasi IR film Ca-Alg/kurkumin/emulsifier menunjukkan bilangan gelombang spesifik pada 3448 cm -1 (gugus hidroksi), 2924 dan 2862 cm -1 (gugus C–H sp 3 emulsifier). Hasil SEM menunjukkan bahwa mikrostruktur permukaan film bervariasi dengan konsentrasi Tween-80 dan Span-80, semakin besar proporsi Tween-80 permukaan film semakin kasar, sedangkan semakin besar proporsi Span-80 permukaan film semakin padat. Laju pelepasan kurkumin selama 8 jam meningkat dari 17,7% menjadi 64,4% ketika komposisi film divariasi dari alginat:Tween-80:Span-80= 74:19:7%(b/b) menjadi 54:41:5%(b/b). Model farmakokinetik menunjukkan bahwa mekanisme pelepasan kurkumin dari alginat mengikuti persamaan Korsmeyer-Peppas yang mengindikasikan mekanisme pelepasan terjadi secara difusi.

The controlled release of curcumin from calcium alginate (Ca-Alg) films that emulsified using Tween-80 and Span-80 had been studied. This research provides an important insight into the effect of hydrophilicity and lipophilicity of the emulsifiers on the release rates of the curcumin from Ca-Alg films. Curcumin-loaded alginate films, which contained different ratio of emulsifiers compositions, have been prepared using calcium chloride (CaCl2) as the cross-linking agent. The films were characterized using Spectrometer IR and SEM (Scanning Electron Microscopy). In vitro controlled release rate study of the curcumin from the films were investigated for 8 hours. Hydrophile-Lipophile Balance (HLB) value of emulsifier Tween-80 and Span 80 was varied in the range of 11,9 to 14,0. The concentration of curcumin that released from the films at various time intervals was determined UV-Vis Spectrophotometrically. IR Spectrometry of Ca-Alg/kurkumin/emulsifier film characterization showed absorption bands at 3448 cm -1 (hydroxyl groups), 1627 cm -1 (carbonyl groups) of the alginate, 2924 and 2862 cm -1 (C–H sp 3 groups of the emulsifier). The result of SEM indicated that the microstructure of the film surface could vary with the concentration of Tween-80 and Span-80. Films for higher concentration of Tween-80 showed rougher surface, but films for higher concentration of Span80 were more compact. Experiment on the release of curcumin showed that the relative amount of curcumin released within 8 hours increased from 17.7% to 64.4% when the film composition was varied from alginate:Tween-80:Span-80= 74:19:7wt% to 54:41:5wt%. The pharmacokinetics model showed that the curcumin release from the alginate film followed Korsmeyer-Peppas equation. This result indicated that the release mechanism was mainly by diffusion process.
