PENGARUH PEMBERIAN EKSTRAK MENGKUDU (Morinda citrifolia L.) “FREEZE JUICING” TERHADAP KADAR GLUKOSA DARAH TIKUS WISTAR DIABETES MELLITUS

Latar belakang : Diabetes Melitus (DM) merupakan penyakit degenerative yaitu kelainan metabolik yang ditandai dengan tingginya kadar glukosa dalam darah. Prevalensi DM terus meningkat dan akan menjadi masalah kesehatan yang serius. Mengkudu merupakan salah satu alternatif pangan fungsional yang memiliki efek hipoglikemik. Mengkudu memiliki bau yang menyengat dan tidak sedap. Metode ektraksi freeze juicing merupakan salah satu alternatif esktraksi sederhana dengan meminimalisir adanya kekurangan tersebut.. Tujuan Penelitian : Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak mengkudu “freeze juicing” terhadap kadar glukosa darah tikus wistar diabetes mellitus. Metode Penelitian : Jenis penelitian ini adalah quasi eksperimental dengan jenis rancangan pre dan post test control group design. Subjek penelitian ini yaitu tiga puluh ekor tikus wistar berumur 2 bulan dengan berat sekitar 160-180 gram. Tikus dibagi dalam 5 kelompok yaitu kontrol negatif, kontrol positif, freeze juicing mengkudu dosis 0,25, dosis 0,50, dan dosis 1 selama 28 hari perlakuan. Tikus diadaptasikan selama 3 hari, kemudian tikus dipuasakan sebelum induksi nicotinamide 110 mg dan streptozotocin 45 mg. Analisis kadar glukosa darah dilakukan sebanyak 2 kali (pre dan post test). Data dianalisis dengan menggunakan ANOVA. Hasil penelitian : Tikus wistar yang diinduksi streptozotocin dan nicotinamide mengalami kenaikan kadar glukosa darah rata-rata >200 mg/dl. Uji statistik yang dilakukan untuk mengetahui perubahan kadar glukosa darah dari pre test ke pos test tidak menunjukkan perbedaan yang signifikan antar kelompoknya dengan p>0,05. Pemberian ekstrak freeze juicing mengkudu tidak berpengaruh terhadap penurunan kadar glukosa darah tikus wistar DM. Walaupun pada kelompok dosis 0.5 mengalami penurunan tetapi pada kelompok kontrol positif juga mengalami penurunan. Kesimpulan : Pemberian ekstrak mengkudu freeze juicing tidak memberi efek pada kadar glukosa darah tikus Wistar diabetes mellitus.

The prevalence of diabetes continues to rise and will be a serious health problem. Noni is an alternative functional foods that have hypoglycemic effects. Noni has a pungent odor and unpleasant. Freeze juicing extraction method is an alternative to the simple extraction to minimize the lack of it. Objective: This study aimed to determine the effect of noni extract \\"freeze juicing\\" on blood glucose levels wistar rats with diabetes mellitus. Method: The type of this research was a quasi experimental study with a type of design form, pre and post test control group design. Subject of this study is thirty Wistar rats aged 2 months and weighs about 160-180 grams. Rats were divided into 5 groups: negative control, positive control, freeze juicing noni doses of 0.25, 0.50 doses, and 1 dose during 28 days of treatment. Rats were adapted for 3 days, then fasted rats before induction of nicotinamide 110 mg and 45 mg streptozotocin. Analysis of blood glucose level was as much as 2 times (pre and post test). Data were analyzed using ANOVA. The results: Wistar rats was induced by streptozotocin and nicotinamide had increase blood glucose levels by an average blood glucose> 200 mg / dl.
