PENGARUH KONSENTRASI EKSTRAK DAUN JAMBU BIJI (Psidium guajava) DALAM ULTRASONIC SCALER COOLANT WATER TERHADAP DAYA HAMBAT PERTUMBUHAN BAKTERI Porphyromonas gingivalis (Kajian in vitro)

Tumbuhan jambu biji (Psidium guajava) merupakan tumbuhan yang banyak tumbuh di Indonesia. Daun jambu biji diduga mempunyai khasiat sebagai agen antibakteri. Tujuan penelitian ini adalah untuk mengetahui konsentrasi ekstrak daun jambu biji (Psidium guajava) dalam ultrasonic scaler coolant water terhadap daya hambat pertumbuhan bakteri Porhyromonas gingivalis. Sebanyak 21 tabung suspensi bakteri disemprotkan ekstrak daun jambu biji (Psidium guajava) konsentrasi 0,009%, 0,019%, 0,039%, 0,078%, 0,156%, akuades sebagai kontrol negatif dan chlorhexidine gluconate 0,0034% sebagai kontrol positif melalui ultrasonic scaler. Larutan suspensi bakteri dan ekstrak diambil 0,2 ml kemudian diratakan di atas plate agar darah menggunakan glass spreader. Selanjutnya, plate agar darah diinkubasi selama 48 jam, suhu 37ËšC dalam inkubator anaerob. Koloni yang terbentuk dihitung oleh 3 pengamat dan dilanjutkan analisis statistik. Hasil uji one way Anova menunjukkan bahwa konsentrasi daun jambu biji dalam ultrasonic scaler coolant water berpengaruh signifikan terhadap daya hambat pertumbuhan bakteri Porphyromonas gingivalis (p<0,05). Hasil uji LSD menunjukkan bahwa terdapat perbedaan yang signifikan antara semua kelompok perlakuan terhadap akuades dan chlorhexidine gluconate 0,0034% (p<0,05) kecuali pada konsentrasi 0,156% terdapat perbedaan yang tidak signifikan terhadap chlorhexidine gluconate 0,0034% (p>0,05). Kesimpulan penelitian ini adalah konsentrasi ekstrak daun jambu biji (Psidium guajava) dalam ultrasonic scaler coolant water berpengaruh terhadap daya hambat pertumbuhan bakteri Porhyromonas gingivalis.

Psidium guajava is a plant that grows in Indonesia. Psidium guajava’s leaf is suspected that have got efficacy as an antibacterial agent. The aim of this study is to determine the effect of concentration of extract of Psidium guajava’s leaf in the ultrasonic scaler coolant water to inhibit growth of Porhyromonas gingivalis. Twenty-one tubes of bacterial suspension were sprayed with an extract of Psidium guajava’s leaf which concentration i.e. 0.009%, 0.019%, 0.039%, 0.078%, 0.156%, distilled water as a negative control and chlorhexidine gluconate 0.0034% as a positive control through ultrasonic scaler. The 0.2 ml of suspension bacterial and extract liquid was taken and was trim on the blood agar plate with a glass spreader. Then, blood agar plates were incubated for 48 hours, with temperature of 37˚C in the anaerobic incubator. Colonies formed were counted by 3 observers and continued with statistical analysis. The results of one way Anova test showed that concentration of Psidium guajava’s leaf in the ultrasonic scaler coolant water has significant effect on the inhibition of bacterial Porphyromonas gingivalis growth (p<0.05). The results of LSD test of showed that there were significant differences between all treatment groups against distilled water and chlorhexidine gluconate 0.0034% (p<0.05), except in the concentration of 0.156%, there was not significant difference against chlorhexidine gluconate 0.0034% (p>0.05). The conclusion of this study is concentration of extract of Psidium guajava’s leaf in the ultrasonic scaler coolant water has got an effect on the inhibition of bacterial Porhyromonas gingivalis growth.
