PENGARUH PEMBERIAN DOSIS EKSTRAK FREEZE JUICING MENGKUDU (Morinda citrifolia) TERHADAP KADAR KOLESTEROL TOTAL DAN HDL-KOLESTEROL PADA TIKUS WISTAR YANG DIINDUKSI DIABETES MELLITUS

Latar Belakang : Penderita penyakit degeneratif semakin lama semakin meningkat. Salah satunya adalah diabetes mellitus. Pada penderita diabetes mellitus sering dijumpai kelainan metabolisme lipid yang dikenal dengan istilah dislipidemia. Salah satu jenis tanaman obat yang dapat menurunkan kadar kolesterol darah adalah mengkudu. Kandungan mengkudu yang dapat menurunkan kadar kolesterol darah adalah senyawa antioksidan antara lain asam ascorbat, ß-caroten, flavonoid, scopoletin. Pengolahan mengkudu dengan freeze juicing dapat menyamarkan aroma tidak sedap pada mengkudu. Tujuan : Mengetahui pengaruh pemberian dosis pada ekstrak mengkudu dengan metode freeze juicing terhadap perbaikan kadar kolesterol total dan HDL, serta mengetahui indeks aterogenik pada tikus diabetes mellitus. Metode : Penelitian ini merupakan penelitian quasi eksperiemental dengan rancangan pre post test melalui control group design. Sebanyak 30 tikus galur wistar jantan berumur 2 bulan. Tikus diadaptasi selama 3 hari, setelah itu diinduksi Streptozotoxin (45 mg/kg BB) dan Nicotinamide (110 mg/kg BB). Setelah tikus menjadi DM, lalu tikus dibagi menjadi 5 kelompok yaitu kontrol negatif (tikus sehat), kontrol positif (tikus DM namun tidak diberi perlakuan), FJ 0,25 (tikus DM dan diberi freeze juicing 0,25), FJ 0,5 (tikus DM dan diberi freeze juicing 0,50), FJ 1,00 (tikus DM dan diberi freeze juicing 1,00). Setelah itu diambil darah untuk dianalisis kadar kolesterol total dan HDL pretest. Setelah 28 hari perlakuan, darah tikus diambil lagi untuk dianalisis kolesterol total dan HDL post test. Hasil : Kadar kolesterol total, HDL, dan indeks aterogenik akhir ditemukan tidak berbeda nyata pada semua kelompok perlakuan (p>0.05) Kesimpulan : Pemberian ekstrak freeze juicing mengkudu tidak memberikan pengaruh terhadap kadar kolesterol total dan indeks aterogenik, namun memberikan pengaruh yang kecil terhadap kadar HDL kolesterol pada tikus wistar yang diabetes mellitus.

Background : Degenerative illness is becoming more intensive now, since a lot of people have a problem with this illness. One of those illness is Diabetes Mellitus. Inside the diabetes sufferer is almost come with lipid metabolism illness known as dislipidemia. One of medication fruit that could be used to cure this illness is mengkudu. The extract of mengkudu that used to decrease blood cholesterol is antioxidant such as asam ascorbat, beta caroten, flavonoid, and scopoletin. By using freeze juicing process it can eliminate stinky smell from mengkudu. Purpose : To understand the effect of mengkudu extract with freeze juicing method for total cholesterol level and HDL. And also to discover aterogenic index inside of wistar mice with diabetes mellitus. Method : this experiment is quasi experimental using pre and postest by control group design. 30 male wistar mouse with all of them in their second month age. Mice were given AIN-93 in 3 days, and then inducted with Streptozotoxin (45 mg/kg of body weight) and Nicotinamide (110 mg/kg of body weight). After mice have diabetes and then they were divided into five groups, named as negatif control (healthy mice), positif control (DM mice with no treatment) FJ 0.25 (DM mice and given freeze juicing 0.25), FJ 0.5 (DM mice and given freeze juicing 0.5), FJ 1.00 (DM mice and given freeze juicing 1.00). Result : The rate of total cholesterol, HDL, and finale index aterogenic were not make any significant difference in all (p>0.05) treatments. Conclution : Adduction of mengkudu’s freeze juicing extract doesn’t make any significant affection onto total cholesterol, from aterogenic index but has a little amount onto HDL cholesterol inside wistar of mice with diabetes mellitus.
