PENGARUH BODY CONDITION SCORE (BCS) TERHADAP KUALITAS SPERMATOZOA SAPI YANG DIKOLEKSI DARI KAUDA EPIDIDIMIS SECARA IN VITRO

2013 | Skripsi | KEDOKTERAN HEWANPenelitian ini dilakukan untuk mengetahui pengaruh Body Condition Score (BCS) terhadap kualitas spermatozoa kauda epididimis yang dikoleksi secara in vitro. Metode BCS merupakan cara yang digunakan untuk menilai kondisi tubuh sapi yang selanjutnya berpengaruh terhadap performa reproduksi sapi. Penelitian dilakukan di Rumah Potong Hewan (RPH) Giwangan. Sampel yang digunakan dalam penelitian ini adalah tiga pasang testis yang berasal dari sapi dengan BCS 2.5, lima pasang testis yang berasal dari sapi dengan BCS 3, dan enam pasang testis yang berasal dari sapi dengan BCS 3.5. Penentuan BCS dengan cara mengamati bagian hook, pin, tailhead, dan rib sapi. Setelah dilakukan penyembelihan kemudian testis dikoleksi. Testis dicuci menggunakan larutan NaCl fisiologis bersuhu 37 o C. Kauda epididimis dipisahkan dari testis kemudian dicacah di cawan petri berisi Phospate Buffer Saline (PBS). Kemudian dilakukan pemeriksaan motilitas spermatozoa dengan mikroskop perbesaran 40x10 dan penghitungan viabilitas spermatozoa dengan pengecatan eosin negrosin. Hasil penelitian menunjukkan bahwa antara berbagai tingkat BCS terdapat perbedaan kualitas spermatozoa meliputi motilitas dan viabilitas spermatozoa. Kualitas spermatozoa sapi dengan BCS 3.5 lebih baik jika dibandingkan dengan sapi dengan BCS 2.5. Dari hasil penelitian disimpulkan bahwa BCS berpengaruh terhadap performa reproduksi sapi.
This study was held to define the effect of Body Condition Score (BCS) on bovine spermatozoa quality that had collected from cauda epididymis by in vitro. That method was done to assess body condition of cows that affect to its reproductive performance. The research was conducted at Rumah Potong Hewan (RPH) Giwangan. Three, five, and six testes were collected from of bovine with BCS 2.5, 3 and 3.5 respectively were used as the material for this studied. First, the determination of hook, pin, tailhead, and ribs were observed. Testes were collected and washed using NaCl Fisiologis 37 o C. In Laboratory, cauda epydidimis were separated from and then sliced using scalpel blade in petri disc consist of Phospat Buffer Saline (PBS). After spermatozoa was released from cauda epydidimis, it was checked the motility with microscope of magnification 40x10 and followed by test viability with eosin-negrosin staining. The result showed that the spermatozoa quality such as motility and viability percentage were difference. Quality of sperm in bovine with BCS 3.5 better than bovine with BCS 2.5. The conclusion of the research were BCS have affected on spermatozoa quality that were colleted from cauda epididymis of bull by in vitro.
