UJI FEKTIVITAS KOMBINASI GLUTARALDEHID-BENZALKONIUM KLORIDA SEBAGAI DISINFEKTAN TERHADAP ISOLAT LAPANG Escherichia coli IN VITRO

Penelitian ini bertujuan untuk mengetahui efikasi disinfektan kombinasi zat aktif glutaraldehid dan benzalkonium klorida terhadap bakteri Escherichia coli dengan melihat konsentrasi hambat minimal dan konsentrasi bakterisidal minimal yang dilakukan in vitro. Sebanyak 8 konsentrasi bertingkat 3,3 ppm; 8,25 ppm; 16,5 ppm; 33 ppm; 49,5 ppm; 66 ppm; 82,5 ppm; dan 99 ppm yang berisi disinfektan dengan zat aktif kombinasi glutaraldehid dan benzalkonium klorida pada media Nutrient broth (NB) diberi isolat Escherichia coli sesuai standar McFarland (108 CFU/mL). Setelah diinkubasi selama 24 jam ditanam pada media agar Eosin Methylen Blue untuk melihat ada atau tidaknya pertumbuhan bakteri Escherichia coli. Hasil menunjukan bahwa kadar hambat minimal kombinasi disinfektan glutaraldehid dan benzalkonium klorida terhadap bakteri Escherichia coli adalah 16,5 ppm, sedangkan kadar bakterisidal minimal adalah 33 ppm.
Escherichia coli contamination in chicken coop can cause colibacilosis that inflicts financial loss for farmers. This study aims to determine the efficacy of a disinfectant that contains the combination of active ingredients glutaraldehyde and benzalkonium chloride against bacteria Escherichia coli by measuring minimum inhibitory concentration and minimum bactericidal concentration performed in vitro. A total of eight serial concentrations 3,3 ppm; 8,25 ppm; 16,5 ppm; 33 ppm; 49,5 ppm; 66 ppm; 82,5 ppm; and 99 ppm which contains the combination of active ingredients disinfectant mixed with glutaraldehyde and benzalkonium chloride on media Nutrient broth (NB) were isolates of Escherichia coli according to McFarland standard (108 CFU/mL). After being incubated for 24 hours,the mixtures of disinfectant and bacteria were cultured on Eosin Methylen Blue agar to see the growth of the bacteria. The result showed that the minimum inhibitory concentration of the combined disinfectant glutaraldehyde and benzalklonium chloride against bacteria Escherichia coli was 16,5 ppm, while the minimum bactericidal concentration was 33 ppm.
