UJI SENSITIVITAS BAKTERI Staphylococcus aureus PADA SUSU MASTITIS SUBKLINIS TERHADAP CEFIXIME DAN PIPERACILLIN DI KELOMPOK TERNAK SARONO MAKMUR WUKIRSARI, CANGKRINGAN, SLEMAN, YOGYAKARTA

2013 | Skripsi | KEDOKTERAN HEWANMastitis merupakan radang pada kelenjar susu, bisa terjadi secara klinis maupun subklinis. Salah satu penyebab mastitis subklinis adalah Staphylococcus aureus. Penelitian ini bertujuan untuk mengetahui sensitivitas Staphylococcus aureus terhadap cefixime dan piperacillin. Sebanyak 16 sampel susu dilakukan uji mastitis dengan reagen CMT. Identifikasi bakteri dilakukan dengan menggunakan media Brain Hearth Infusion (BHI), Plat Agar Darah (PAD), dan MacConkey Agar (MCA). Pengujian biokimia, pengecatan Gram, uji katalase, uji koagulase, dan uji DNase dilakukan untuk peneguhan jenis bakteri yang tumbuh. Uji sensitivitas bakteri dilakukan pada Muller Hinton Agar (MHA), dan diberi antibiotik dengan menggunakan paper disc. Zona hambatan diukur setelah media diinkubasi 24 jam dengan suhu 37 o C. Hasil isolasi menunjukkan 2 dari 16 sampel (12,5 %) dinyatakan positif terinfeksi Staphylococcus aureus. Sensitivitas Staphylococcus aureus terhadap piperacillin 100 % sedangkan terhadap cefixime bakteri ini 100 % resisten.
Mastitis is an inflammation in the mammary gland. It may infect clinically or subclinically. One of the causal factors of subclinical mastitis is Staphylococcus aureus. This study aims to determine the sensitivity level of Staphylococcus aureus toward Cefixime and Piperacillin. Sixteen samples of milk have been through mastitis test using reagent CMT. The samples were inoculated in BHI and then were planted in blood plate and MacConkey Agar (MCA) with the incubation period was 24 hours in the temperature of 37 o C. The grown colonies were grouped based on the morphology. Biochemic test, Gram’s staining, catalase test, coagulase test, and DNase test were conducted to confirm what kind of bacteria that grew. To figure out the sensitivity level of the bacteria, the colonies growing on NA medium were diluted with NaCl, planted on Muller Hinton Agar (MHA), and then given the antibiotics using paper disc. The zone of inhibition was measured after the media was incubated for 24 hours in the temperature of 37 o C. The result of the isolation showed 2 of 16 samples (12.5%) were positive infected by Staphylococcus aureus. It can be concluded that the sensitivity level of Staphylococcus aureus toward piperacillin is 100% while its resistance level toward cefixime is 100%.
