JUMLAH SPERMATOSIT PRIMER DAN KADAR TESTOSTERON INTRATESTIKULER PADA TIKUS Sprague Dawley YANG DIINDUKSI STREPTOZOTOCIN DENGAN LATIHAN TERATUR TERUKUR

2013 | Tesis | S2 Ilmu Kedokteran Dasar dan BiomedisLatarBelakang :Pada penderita diabetes mellitus terjadi hiperglikemia yang melatar belakangi peningkatan stress oksidatif dan pembentukkan Advance Glycation Endproduct. Kedua hal ini berperan penting dalam komplikasi kronis diabetes mellitus pada organ reproduksi testis. Follicle Stimulating Hormon akan menstimulus sel Sertoli untuk proses spermatogenesis; Luteinizing Hormon akan mendorong sel Leydig dalam menghasilkan hormontestosteron bagi proses spermatogenesis; dan Growth Hormon/Insulin-like Growth Factor-1 merupakan factor autocrin/paracrin dalam mengatur jumlah dan fungsi sel Leydig dewasa, sebagai stimulator proliferasi dan factor mitogenik langsung dalam menstimulasi aktivitas proliferasi sel germinal. Tujuan :Tujuan penelitian ini adalah membuktikan latihan teratur terukur dalam memberikan perubahan dalam jumlah spermatosit primer dan kadar testosteron intratestikuler pada testis tikus Sprague dawleyyang diinduksi STZ. Secara detail mengukur lapisan sel germinal dan diameter tubulus seminiferous, menghitung jumlah spermatosit primer, mengukur kadar testosterone intratestikuler, serta menghitung kekuatan korelasi jumlah spermatosit primer dengan kadar testosteron intratestikuler. Metoda :Penelitian ini merupakan kuasieksperimental dengan post test only control group design. Subyek adalah 15ekor tikus Sprague Dawley yang dibagi secara acak menjadi 3kelompok. Kelompok normal tidak mendapat induksi streptozotocin dan tanpa perlakuan latihan teratur terukur. Kelompok kontrol mendapat induksi streptozotocin 35 mg/kgbb dan tanpa perlakuan latihan teratur terukur.Kelompok perlakuan mendapat induksi streptozotocin 35 mg/kgbb dan mendapatkan latihan teratur terukur selama 9 minggu. 5 minggu pertama latihan teratur terukur intensitas meningkat dari 5 meter/menit selama 10 menit sampai 17 meter/menit selama 45 menit. 4 minggu terakhir intensitas konstan 20 meter/menit selama 60 menit. Setelah terminasi tikus, 1 testis dibuat sediaan preparathistologi testis dengan pewarnaan PASdantestis lain dihomogenisasi untuk diukur kadar testosteronintratestikuler dengan metoda ELISA. Uji statistik dengan one way ANOVA dan uji LSD multiple comparison. Hasil: Jumlah spermatosit primer pada kelompok perlakuan lebih besar secara signifikan dibandingkan kelompok kontrol (p<0,05). Lapisansel germinal dan diameter tubulusseminiferus, serta kadar testosteronintratestikuler lebih besar tidak signifikan pada kelompok perlakuan dibandingkan kelompok kontrol. Jumlah spermatosit primer tidak menunjukan korelasi yang kuat dengan kadar testosteron intratestikuler (p=0,49; r=0,201). Kesimpulan:Latihan teratur terukur memberikan efek terhadap jumlah spermatosit primer, namun tidak memberikan efek terhadap kadar testosteron intratestikuler tikus Sprague dawley yang diinduksi streptozotocin. Jumlah spermatosit primer tidak menunjukan korelasi yang kuat dengan kadar testosteron intratestikuler.
Background :Hyperglycemia often happens in people with diabetesmellitus. It affects on the increase of oxidative stress and the formation of advanceglycationend products. Both have an important role in the chronic complication on the reproductive organs : the testicle. Follicle Stimulating Hormone will stimulate Sertolicells for the spermatogenesis process, while Luteinizing Hormone will stimulate Leydig cells to produce testosterone hormone for the process of spermatogenesis. Growth Hormone/Insulin-like Growth Factor-1 is anautocrin/paracrinfactor in regulating the number and function of the mature Leydig cells. Growth Hormone/Insulin – like Growth Factor-1 serves as the proliferation stimulator and directmitogenic factor in stimulating the germcells proliferation. Objectives :This research is to study the effects of measured regular-exercise in changing the number of primary spermatocyte and the level of intratesticular testosterone in the testicles of Sprague dawleyrats induced by streptozocin. The research counts the number of primaryspermatocyte, the level of intratesticular testosterone and the correlation between the number of primary spermatocyte and the level of intratesticular testosterone. In detail, this also measures the germ cell layers and the diameter of seminiferous tubules. Methods :This research is a quash experimentwith a control group design on post test only. The subjects are 15 Sprague dawleyrats divided randomly into 3 groups. First group is a normal one. They do not get any streptozocin inductions and measured regular-exercise treatment. The second or the control group receives streptozocin inductions of 35 mg/kg body weight but they do not get any measured regular-exercise treatment. The third or the treatment group gets streptozocin inductions of 35 mg/kg body weight and measured regular-exercise treatment for 9 weeks. During the first 5 weeks, the intensity of the exercise is increased from 5 m/minute for 10 minutes to 17m/minute for 45 minutes. In the last 4 weeks, this group gets a constant intensity of the exercise that is 20m/minute for 60 minutes. After the exercise ends/ terminates, 1 testis is made for the preparat preparation for testicle histology by fitting staining and the other is homogenized to check its intratesticular testosterone level using ELISA method. The statistics is evaluated by One-way ANOVA test and multiple comparison of LSD Test. Results :The number of primary spermatocyte in the treatment group is significantly larger than the control group (p < 0.05). The germ cell layers, the diameter of seminiferous tubules and testosterone intratesticularlevels in this group are not larger significantly than the control group.The number of primary spermatocyte is correlated insignificantly to the intratesticular testosterone levels. (p = 0.49; r = 0.201). Conclusion : Based on the research, measured regular-exercise has just affected on the primary spermatocyte number, but it does not give any effects on the intratesticular testosterone level Sprague dawleyrats induced by streptozocin. The primary spermatocyte number does not correlate strongly with the intratesticular testosterone level, too.
