POTENSI PEPTIDA BIOAKTIF DARI EKSTRAK RANGGAH VELVET RUSA SAMBAR (Cervus unicolor brookei) SEBAGAI AGEN ANTIHIPERTENSI

Angiotensin converting enzyme (ACE) memegang peranan penting dalam pengaturan tekanan darah dalam sistem renin-angiotensin. Protein ranggah velvet rusa Sambar (Cervus unicolor brookei) terdiri dari 14 macam asam amino, diduga dapat dijadikan sebagai sumber peptida bioaktif yang dapat digunakan sebagai agen antihipertensi. Tahapan penelitian yang dilakukan yaitu : preparasi protein ranggah velvet, digesti protein ranggah velvet menggunakan pepsin dan tripsin, pengujian aktivitas peptida penghambat ACE dan purifikasi peptida penghambat ACE. Monitoring proses digesti protein ranggah velvet menggunakan beberapa parameter yaitu : konsentrasi protein terlarut, peptide content dan profil berat molekul fraksi-fraksi protein (native protein, hasil digesta menggunakan pepsin dan hasil digesta menggunakan tripsin). Konsentrasi protein terlarut (mg/ml) diukur menggunakan uji Biuret pada native protein adalah 19,05 ± 0,62, sedangkan setelah diberi pepsin dan tripsin adalah 22,22 ± 0,89 dan 16,03 ± 0,84 secara berurutan. Peptide content (mg/ml) pada native protein, digesta pesin dan digesta tripsin berturut-turut adalah 2,040 ± 0,004; 2,080 ± 0,006 dan 2,160 ± 0,003. Berat molekul (dalton) ranggah velvet berturut-turut adalah 53.932,80; 33.424,03 dan 16.306,76. Aktivitas penghambatan (inhibition activity) hidrolisat protein ranggah velvet setelah purifikasi menggunakan SEP-PAK Plus C-18 Catridge terhadap ACE adalah sebesar 80,37% dan Nilai IC50 sebesar 0,433 mg/ml.
Hypertension is the most common serious chronic health problem in adults and carries a high risk factor for arteriosclerosis, strokes, and end-stage renal disease. Hypertensive patients can control their blood pressure by taking medicine to inhibit angiotensin converting enzyme (ACE). ACE plays an important role in regulation of blood pressure in the renin-angiotensin system. Protein of velvet antler of Sambar deer (Cervus unicolor brookei) contains 14 amino acids expected as a source of bioactive peptides that can be used as antihypertensive agent. The stage of this research were : preparation of velvet antler protein, digestion of velvet antler protein by pepsin and trypsin, determination of ACE inhibitory activity, and purification of ACE inhibitory peptides. Digestion process monitoring used some parameters, that were : concentration of soluble protein, peptide content, and profile of molecular weight of native protein fraction as well as after pepsin and trypsin digestion. The result showed concentration of soluble protein (mg/ml) using biuret test was 19.05 ± 0.62 while after pepsin and trypsin were 22.22 ± 0.89 and 16.03 ± 0.84 respectively. Peptide content (mg/ml) of velvet antler was 2.040 ± 0.004 while after pepsin and trypsin were 2.080 ± 0.006 and 2.160 ± 0.003 respectively. Molecular weight (daltons) of velvet antler consecutive was 53,932.80; 33,424.03 and 16,306.76. Inhibitory activity of velvet antler protein on ACE was 83.38% and the IC50 value was 14.512 mg/ml. Inhibitory activity velvet anthler protein hydrolyzed after purification using SEP-PAK Plus C 18 Cartridge on ACE was equal to 80,37% and the IC50 value was 0.433 mg/ml
