IsolataktifdaunT. diversifolia(TD) bersifatselektif terhadap sel kanker kolon (WiDr) dengan IC50= 0,585danIndeksSelektivitas= 69, 015. Tujuan Penelitian: Mengetahui potensi isolataktifdaun TD sebagai antiproliferasi melalui pengamatan siklus sel WiDrdanpotensi antiangiogenesisnya melalui pengamatan ekspresi VEGF. Metode Penelitian: Isolasi TD dilakukan dengan metode KLT preparatif. Analisis siklus sel WiDr dengan metode flowcytometry menggunakan propidium iodide (PI).Analisis antiangiogenesis dengan metode immunositokimia menggunakan human antibodi monoklonal anti VEGF. Hasil:Isolataktifdaridaun TD dosis4 µg/mL menghambatsiklusselpadafase S (jam ke-24,36,dan 72) danfase G1 (jam ke-48). Padadosis 8 µg/ml menghambat G2/M (jam ke-24), subG1 (jam ke-36 dan 48), danfase S (jam ke-72). Ekspresi VEGF secarabermaknaturundari 6,29% menjadi 3,31% padaperlakuanisolataktif TD dosis 4 µg/mL. Kesimpulan:IsolataktifdaridaunTD padadosis4 µg/mL and 8 µg/mL menghambatsiklusselpadafasenonspesifikdanmenurunkanekspresi VEGF padadosis 4 µg/mL.
Background: Colon cancer in Indonesia is still ranked in the 10 primary malignant tumor diseases of inpatients at the hospital. New approaches to prevent and treat colon cancer has developed to more selective. Cell cycle and angiogenesis play an important role in tumor progression and metastasis then those contribute to patient prognosis.Active compound isolated from the leaves of T. diversifolia (TD) is most selective on WiDr cell line with IC50 = 0.585 ug/ml and selectivity index = 69, 015. Objective: To know the antiproliferative and antiangiogenesis potency of the active compound isolated from the leaves of TD through observation on WiDr cell cycle and VEGF expression. Methods: Isolation of TD is performed by preparative TLC method. WiDr cell cycle is analyzed by flow cytometry using propidium iodide (PI). Analysis of antiangiogenesis with immunocytochemistry method using anti-VEGF human monoclonal antibody. Results : Active compound isolated from the leaves of TD at 4 µg/mL inhibit the cell cycle at S (24,36, and 72 hours) and G1 phase (48 hours). It dose at 8 µg/ml inhibit at G2/M (24 hours), subG1 (36 and 48 hours), and S phase (72 hours). The expression of VEGF is significantly depleted from 6,29% (negative control group) to become 3,31% at 4 µg/mL. Conclusion: Active compound isolated from the leaves of TD at 4 µg/mL and 8 µg/ml inhibit the cell cycle in nonspecific phase and at 4 µg/ml deplete the VEGF expression on human colorectal cancer WiDr cells.
