PENGARUH FRAKSI TAK LARUT HEKSAN EKSTRAK ETANOLIK BUAH CABE JAWA (Piper retrofactum Vahl) TERHADAP EFEK EKSTRAK ETANOLIK AKAR PASAK BUMI (Eurycoma longifolia Jack) PADA AKTIVITAS SEKSUAL DAN KADAR TESTOSTERON TIKUS JANTAN WISTAR

Penelitian terhadap akar pasak bumi dan cabe jawa sebagai afrodisiaka telah banyak dilakukan. Penelitian ini bertujuan untuk mengetahui pengaruh fraksi tak larut heksan ekstrak etanolik buah cabe jawa terhadap efek ekstrak etanolik akar pasak bumi pada aktivitas seksual tikus jantan Wistar dengan parameter intromission, climbing, dan coitus. Juga untuk mengetahui pengaruh pemberian kombinasi fraksi tak larut heksan ekstrak etanolik buah cabe jawa dan ekstrak etanolik akar pasak bumi terhadap peningkatan kadar testosteron tikus jantan Wistar. Perlakuan per oral sekali sehari selama 30 hari terdiri dari kombinasi fraksi tak larut heksan ekstrak etanolik buah cabe jawa dengan dosis 31,72 mg, 63,4 mg, dan 126,88 mg/kgBB dengan ekstrak etanolik akar pasak bumi 500 mg/kgBB (K1, K2, K3), ekstrak etanolik akar pasak bumi 500 mg/kgBB (EPB), dan fraksi tak larut heksan ekstrak etanolik buah cabe jawa 126,88 mg/kg BB (FTLH). Pengamatan aktivitas seksual dilakukan selama 60 menit 2 hari sekali di minggu pertama dan empat hari sekali di minggu berikutnya. Kadar testosteron ditentukan pada hari ke-31. Nilai rata-rata luas AUC introduction, climbing, coitus dan ratarata kadar testosteron dianalisa statistik dengan tingkat kepercayaan 95%. Hasil penelitian menunjukkan bahwa perlakuan K1, K2 dan K3 tidak memberikan perbedaan yang bermakna terhadap peningkatan aktivitas seksual dibandingkan dengan EPB (p>0,05) pada semua parameter. Demikian juga pada pengujian kadar testosteron tidak memberikan perbedaan yang bermakna dalam meningkatkan kadar testosteron dibandingkan dengan andriol sebagai kontrol positif ataupun CMC natrium sebagai kontrol negatif. Berdasarkan hasil penelitian, disimpulkan bahwa penambahan fraksi tak larut heksan ekstrak buah cabe jawa pada ekstrak pasak bumi 500 mg/kg/BB tidak berpengaruh terhadap peningkatan efek ekstrak etanolik akar pasak bumi pada aktivitas seksual tikus jantan Wistar dan tidak meningkatkan kadar testosteron tikus jantan Wistar.

Researches on the pasak bumi roots and cabe jawa fruit as aphrodisiac has done. The aim of the research is to determine the effect of insoluble hexane fraction of ethanolic extract of cabe jawa fruit to effect of ethanolic extract of pasak bumi root on sexual activity of Wistar male rats with the parameters of intromission, climbing, and coitus. Also to determine the effect of the combination of insoluble hexane fraction of ethanolic extract of cabe jawa fruit and ethanolic extract of pasak bumi root to the elevated testosterone levels in Wistar male rats. Treatment orally once daily for 30 days consisted of a combination insoluble hexane fraction of ethanolic extract of cabe jawa fruit 31.72 mg; 63.44 mg; 126.88 mg/kg and ethanolic extract of pasak bumi root 500 mg/kg (K1, K2, K3); ethanolic extract of pasak bumi root 500 mg/kg (EPB); insoluble hexane fraction of ethanolic extract of cabe jawa fruit 126,88 mg/kg (FTLH). The observation of sexual activity performed for 60 minutes once in two days on the first week and once in four days on the next weeks. Testosterone levels were determined on the 31st day. The average value of AUC of introduction, climbing, coitus and the average levels of testosterone were analyzed statistically with confidence level at 95%. The results showed that treatment with K1, K2 and K3 have no significant differences to the increased of sexual activity compared to the EPB (p>0.05) on all parameters. Similarly on testing testosterone levels, no significant difference in increasing testosterone levels compared with andriol as positive control and sodium CMC as negative control. Based on the results of research, concluded that the addition of insoluble hexane fraction of ethanolic extract of cabe jawa fruit to etanolik extract of pasak bumi root in dosage 500 mg/kg have no affect on the increased of sexual activity effect of the ethanolic extract of pasak bumi root and no increase testosterone levels in male rats Wistar.
