PENGARUH PEMBERIAN CAMPURAN MADU, JINTAN HITAM, PROPOLIS DAN ROYAL JELI TERHADAP SEL -PANKREAS DAN KADAR MALONDIALDEHID PADA TIKUS WISTAR PASCA PAPARAN STREPTOZOTOCI

Latar Belakang : Hiperglikemi kronis dapat menimbulkan radikal bebas. Radikal bebas menyebabkan peningkatan stres oksidatif dan kenaikkan peroksidasi lipid serta kerusakan sel β pankreas. Hasil akhir peroksidasi lipid adalah malondialdehid (MDA). Kerusakan sel β pankreas menurunkan produksi insulin sehingga menjadi hiperglikemi kronis. Aktivitas antioksidan yang terdapat pada madu, jinten hitam, propolis dan royal jeli diharapkan dapat menurunkan radikal bebas, sehingga dapat menurunkan kadar MDA dan jumlah vakuola sel β pankreas. Tujuan Penelitian : Untuk mengetahui pengaruh campuran madu 40%, jinten hitam 30%, propolis 20% dan royal jeli 10% terhadap kadar MDA dan jumlah vakuola sel β pankreas. Metode penelitian : Tiga puluh ekor tikus putih galur Wistar jantan dibuat hiperglikemia dengan streptozotocin secara intraperitonial (kelompok 2, 3, 4, 5 dan 6) dan enam ekor tikus tetap dibuat normal. . Kelompok 4, 5, 6 kemudian diberi perlakuan sediaan kombinasi madu 40%, jintan 30%, propolis 20% dan royal jeli 10% dosis 0,6; 1,2; 2,4 ml/200gBB. Kelompok 3 merupakan kelompok kontrol (+) yang diberi metformin 9 mg/200gBB, kelompok 2 merupakan kelompok kontrol (-) diberi air masak dan kelompok 1 merupakan kelompok normal yang tidak hiperglikemia. Kadar MDA diukur pada hari ke-0, 3, 10 dan 17 dan jumlah vakola sel β pankreas pada hari ke-17. Hasil : Dari penelitian ini didapatkan rata-rata kadar MDA kelompok 3, 4, 5 dan 6 mengalami penurunan, sedangkan kelompok 2 mengalami kenaikkan dan kelompok 1 relatif stabil. Dari hasil analisis one way ANOVA pada hari ke-17 menunjukkan penurunan yang sebanding dan tidak ada perbedaan antara kelompok 6 dengan kelompok metformin. Sedang jumlah vakuola pada hari ke-17 paling rendah pada kelompok 4 yaitu pemberian campuran dengan dosis 0,6 ml / 200g BB. Kesimpulan : Pemberian campuran madu 40%, jintan hitam 30%, propolis 20% dan royal jeli 10% selama 14 hari dengan dosis 1,2 dan 2,4 ml / 200 g BB dapat menurunkan jumlah vakuola sel β pankreas yang tidak berbeda dengan pemberian metformin. Dosis 0,6 ml / 200 g BB menurunkan lebih banyak dibanding metformin. Pemberian campuran madu 40%, jintan hitam 30%, propolis 20% dan royal jeli 10% selama 14 hari dengan dosis 0,6 dan1,2 ml / 200 g BB menurunkan kadar MDA yang lebih kecil dibanding dengan pemberian metformin (9 mg / 200g BB). Campuran dosis 2,4 dapat menurunkan kadar MDA yang tidak berbeda dengan metformin (9 mg / 200 g BB).

Research Background: Hyperglycemic chronic could cause free radical. Free radical caused an increasing of oxidative stress, reducing the antioxidant, and an increasing of peroxidative lipid, and the damage of β pancreas cell. The final product of peroxidative lipid was malondialdehid (MDA). The damage of β pancreas cell decreased the insulin production so that it caused hyperglycemia chronic. The anti-oxidant activities of honey, black dill, propolis, and prodigal broodinging were expected to reduce free radical, so that they could reduce the MDA level and the amount of β pancreas cell vacuole. Research Objectives: to indicate the influence of the mixture of 40% honey, 30% dill, 20% propolis, and 10% prodigal broodinging to the MDA level and the amount of β pancreas cell vacuole. Research Method: Thirty white male Wistar hyperglycemic rat intraperitonially inducted with STZ (Group 2 – 6) and six normal rats. Groups 4, 5, and 6 were treated with the mixture of honey, black dill, propolis, and prodigal broodinging with the dosage 0,6; 1,2; 2,4 ml/200gr weight. Group 3, was a positive control group, was treated with of metformin 9mg/200gr weight, group 2, was a negative control group, was treated with boiled water 2ml/200gr weight. Group 1 was a normal group, was not inducted with STZ. The MDA level was scaled on the day 0, 10th, and 17th. The vacule of β pancreas cell was calculated on the day 17th. Results: Based on this research it found that the average of MDA level of group 3, 4, 5, and 6 were decreasing, meanwhile the average MDA level of group 2 was increasing and was normal in group 1. Based on the analysis One Way Anova on the day 10th, it showed that there were decreasing level from the groups with minimal treatment than the group without metformin. On the day 17th, there were the same decreasing and there was no difference among group 6 and metformin group. Meanwhile, the group 4, which treated with the mixture with dosage 0,6 ml/200gr weight, had the lowest amount of vacuole on day 17th. Conclusion: The amount of β pancreas cell vacuole of hyperglikemic rat with the mixture of 40% honey, 30% dill, 20% propolis, and 10% prodigal broodinging for 14 days dosage 1.2 and 2.4 ml/200gr weight had not significant differences with hyperglycemic rat with metphormin 9 mg /200mg weight, however the hyperglycemic rat with the dosage mixture 0,6 ml/200gr weight could reduce more β pancreas cell vacuole than the rat with metformin. The mixture of 40% honey, 30% dill, 20% propolis, and 10% prodigal broodinging for 14 days dosage1.2 and 2.4 ml/200gr weight could reduce lower MDA level than the rat with metformin and the MDA level of hyperglycemic rat with the dosage mixture 0,6 ml/200gr weight and the rat with metformin had not significant difference.
