2011 | Tesis | S2 Ilmu Kedokteran Dasar dan BiomedisKurkumin merupakan zat aktif yang terdapat dalam Curcuma longa. Kurkumin memiliki banyak aktivitas biologik , salah satunya adalah anti fertilitas. Penelitian kurkumin pada sistem reproduksi pria telah dilakukan. Kurkumin menurunkan berat testis dan testosteron darah. Penelitian ini dilakukan untuk mengetahui efek kurkumin terhadap kadar testosteron intratestikular tikus dan mengetahui letak kerja kurkumin dalam jalur transduksi sinyal steroidogenesis di sel Leydig. Penelitian dilakukan pada 27 ekor tikus Sprague dawley jantan usia 90 hari. Hewan coba dibadi menjadi 9 kelompok perlakuan yaitu kelompok 1 diberi akuades, kelompok 2 diberi hCG, kelompok 3 diberi CMC, kelompok 4 diberi kurkumin, kelompok 5 diberi hCG, kurkumin, kelompok 6 diberi teofilin, kelompok 7 diberi hCG dan teofilin, kelompok 8 diberi teofilin dan kurkumin, dan kelompok 9 diberi hCG, teofilin dan kurkumin. Tikus dikorbankan pada saat umur 91 hari. Testis diambil dan dihomogenizer lalu disentrifus. Supernatan diambil untuk diukur kadar testosteron intratestikular. Hasil penelitian menunjukkan bahwa kadar testosteron intratestikular (ng/ml) dari 9 kelompok perlakuan, masing-masing n = 3, adalah (1) 44.36±1.30 (akuades), (2) 41.53±16.44 (hCG), (3) 37.33±9.05 (CMC), (4) 54.16±29.82 (kurkumin), (5) 49.36±5.05 (hCG + kurkumin), (6) 44.20±22.09 (teofilin), (7) 83.23±18.81 (hCG + teofilin), (8) 44.73±22.90 (teofilin + kurkumin), (9) 47.70±16.47 (hCG + teofilin + kurkumin). Analisa statisik dengan ANOVA menunjukkan tidak ada perbedaan bermakna (p>0,05). Analisa statistik dengan planned comparison with contrast didapatkan perbedaan bermakna antara kelompok K7 dengan K2 dan K9 (p<0,05). Kesimpulan penelitian ini adalah kurkumin tidak menurunkan kadar testosteron intratestikular tikus dan letak kerja kurkumin berada di sebelum cAMP pada transduksi sinyal steroidogenesis sel Leydig.
Curcumin is an active agent on Curcuma longa. Curcumin had so many biologic activities, one of them is anti fertility. Curcumin research in male reproductive system had been done. Curcumin reduced testis weight and serum testosterone. This study is done to find out site action of curcumin in signal transduction pathway steroidogenesis on Leydig cell and to know curcumin effect on testosterone intratesticular level in rats. This study was conducted to 27 male Sprague dawley rats aged 90 days. Rats divided into 9 groups treatment : group 1 rats were given aquades, group 2 rats were given a single dose of 50 IU hCG, group 3 rats were given CMC, group 4 rats were given curcumin, group 5 rats were given hCG and curcumin, group 6 rats were given theophylline, group 7 rats were given hCG and theophylline, group 8 rats were given theophylline and curcumin, group 9 rats were given hCG and theophylline and curcumin. Rats were sacrified on age 91 days. Testis was taken, homogenizer and sentrifuge. Supernatant was taken in order to measured intratesticular testosterone level. The result of this study show intratesticular testosterone level from 9 group treatment, each group are n = 3, are (1) 44.36±1.30 (aquades), (2) 41.53±16.44 (hCG), (3) 37.33±9.05 (CMC), (4) 54.16±29.82 (curcumin), (5) 49.36±5.05 (hCG + curcumin), (6) 44.20±22.09 (theophylline), (7) 83.23±18.81 (hCG + theophylline), (8) 44.73±22.90 (theophylline + curcumin), (9) 47.70±16.47 (hCG + theophylline + curcumin). Statistic analytic with ANOVA showed there is no significant difference (p>0.05). Statistic analytic with planned comparison with contrast showed there is significant difference on K7 with K2 and K9 group (p<0,05). This study have a conclusion that curcumin not decreased testosterone intratestikular level in rats and site action of curcumin located on before cAMP in signal transduction pathway steroidogenesis on Leydig cell .
